Summary
In guinea pigs the toxicity of k-strophanthine given by i.v. infusion is diminished by pretreatment with the adrenergic blocking agent Kö 592 as well as by iproveratril. With both substances the frequency of the heart is diminished, toxic changes of the ECG are retarded and the lethal dose is elevated.
In isolated spontaneously beating auricles of the guinea pig the blocking agent propranolol inhibits similar to quinidine the toxic effect of k-strophanthine whereas the positiv inotrop effect of k-strophanthine is unaffected.
In isolated electrically driven atria the toxic effect of k-strophanthine is not affected by pretreatment with propranolol and quinidine. Also pretreatment with reserpine does not alter the effect of k-strophanthine in the isolated auricle preparation.
Our experiments indicate that the effect of k-strophanthine in the isolated heart preparation is independent of the cathecholamine content of the heart and that the antagonism of the adrenergic blocking agent is related to their quinidine-like action.
Zusammenfassung
Bei der Titration an Meerschweinchen wird die Toxicität des k-Strophanthins durch den β-Receptorenblocker Kö 592 sowie durch Iproveratril vermindert. Beide Substanzen haben eine bradykarde Eigenwirkung, verzögern das Auftreten toxischer EKG-Veränderungen und erhöhen die Dosis letalis.
Am isolierten, spontanschlagenden Vorhofpräparat des Meerschweinchens werden toxische k-Strophanthinwirkungen durch den β-Receptorenblocker Propranolol ähnlich wie durch Chinidin gehemmt, während die positiv inotrope Wirkung des k-Strophanthins erhalten bleibt.
Am elektrisch gereizten Vorhofpräparat wird die toxische Wirkung des k-Strophanthins durch Propranolol oder Chinidin nicht vermindert. Auch am Vorhofpräparat Reserpin-behandelter Tiere bleibt die Wirkung des Strophanthins unverändert. — Die Versuche sprechen dafür, daß die Wirkung des k-Strophanthins am isolierten Vorhofpräparat ohne Beteiligung der Brenzcatechinamine zustande kommt und der Antagonismus des β-Receptorenblockers auf einer Chinidin-ähnlichen Wirkung beruht.
