Summary
1. In suspensions of human erythrocytes, the levels of reduced NAD and NADP were lowered to about 30% by phenylhydroxylamine in a concentration of 5 · 10−4 M. Phenylhydroxylamine was effective under anaerobic and aerobic conditions. The phenylhydroxylamine action was related to phenylhydroxylamine concentrations and time of incubation.
2. Concentrations of phenylhydroxylamine higher than 10−3 M led to an additional decrease of NAD and NADP levels in erythrocytes.
3. Besides this action, indirect influences of phenylhydroxylamine also diminish the reduced pyridinenucleotide concentrations in erythrocytes. In a cycling process, NADPH dependent enzymes reduce the nitrosobenzene formed from phenylhydroxylamine in erythrocytes to phenylhydroxylamine. NADPH enzymes also reduce GSSG that is additionally oxidized from GSH by phenylhydroxylamine.
On the other side, NADH is involved in the NADH-linked methaemoglobin reduction.
