Summary
In this work an attempt was made to examine the active concentration mechanism of radioiodide in the trachea of guinea pigs by the histo-autoradiographic localization of 131J− (125J−), 35SCN−, 36Cl− and 58Co2+ in vivo and in vitro.
The active concentration of radioiodide takes place in both directions most probably in special cells in layers of the tracheal epithel which are nearest to the lumen in vitro and in vivo: Lumen →← epithelial cell. It will be discussed which of the three epithelial cell types (cilia, brush or goblet cell) participates in this active process.
In vitro the active concentration mechanism of those cells is disturbed by atropine sulfate, KCN, NaF, 2,4-dinitrophenole, monoiodacetate as well as by longer incubation in a Ringer-phosphate-glucose-medium. Under these conditions a diffusion of radioiodide through the trachea probably due to the damage of the metabolism of the cells can be seen. Radioiodide and 35S-labeled thiocyanate are autoradiographically localized in the cytoplasma and intercellular in the tracheal epithel of guinea pigs. From this the reciprocal displacement of both anions is concluded.
In vitro 35SCN− and 36Cl− are localized equally in the cytoplasma of epithelial cells of the guinea pig trachea. In vivo the perichondrium of tracheal cartilage enriches 35SCN−, in vitro it does not. In the gl. tracheales radioiodide, 35SCN− and 36Cl− is localized in the cytoplasma of secreting cells as well as in the lumen. 58Co2+ is only absorbed on the surface of epithelial cells.
The results are discussed in relation to the different autoradiographic techniques (dry-stripping, alcohol-stripping, reversed-stripping of frozen dried sections and paraffin sections) practised in this work. The localization of water soluble radioions in different tissues is ascertained if different histo-autoradiographical methods show separately the same results.
Zusammenfassung
Mit Hilfe der autoradiographischen Lokalisation von 131J-bzw. 125J−, 35SCN−, 36Cl− und 58Co2+ in vivo und in vitro wird versucht, Einblicke in die aktive Anreicherung von Radiojodid in der Trachea des Meerschweinchens zu bekommen. Die aktive Anreicherung von Radiojodid erfolgt mit großer Wahrscheinlichkeit in bestimmten Zellen der lumennahen Schicht des Trachealepithels in vivo und in vitro in beiden Richtungen Lumen ⇄ Epithel. Welche von den drei Epithelzelltypen (Cilien-, Bürsten- oder Schleimzelle) für diesen aktiven Prozeß in Frage kommt, wird diskutiert.
In vitro wird der aktive Anreicherungsprozeß in diesen Zellen durch Stoffwechselgifte (Atropinsulfat, KCN, NaF, 2,4-Dinitrophenol, Monojodacetat) und bereits längere Inkubation in einem Ringer-Phosphat-Glucose-Medium gestört. Die hierbei beobachtete Diffusion von Radiojodid durch die Trachea wird auf eine Schädigung des Zell- bzw. des Gewebestoffwechsels durch jene Bedingungen zurückgeführt.
Radiojodid und 35SCN− sind in vivo in der MUcosa der Meerschweinchentrachea in gleicher Weise lokalisiert: Im Cytoplasma der lumennahen Epithelzellen und intercellulär. Hieraus läßt sich die gegenseitige Verdrängung beider Ionen ableiten.
In vitro zeigen 35SCN− und 36Cl− ähnlich lokalisierte Schwärzungen in der Meerschweinchentrachea (Cytoplasma von Epithel- und Drüsenzellen). In vivo wird 35SCN− im Perichondrium des Knorpels angereichert, in vitro nicht. Schwärzungen durch Radiojodid, 35SCN− und 36Cl− werden über dem Cytoplasma der Drüsenzellen sowie im Lumen der Glandulae tracheales gefunden. 58Co2+ wird in vitro von der Trachea nicht aufgenommen: Schwärzungen durch Radiocobalt werden nur im Bereich der Cilien der Epithelzellen gesehen.
Die Ergebnisse werden anhand der verschiedenen autoradiographischen Techniken (Trocken-Stripping, Alkohol-Stripping, Umkehr-Stripping von Gefrierund Paraffinschnitten) diskutiert und festgestellt, daß bei den hier angewandten Methoden ein sicherer Hinweis für die Lokalisation eines wasserlöslichen Radionukleids nur dann besteht, wenn verschiedene, voneinander unabhängige Methoden das gleiche Ergebnis bringen.
