The effect of paraoxone on the electrical activity of the preganglionic sympathetic nerve and on the blood pressure in rats

Summary

The effects of a cholinesterase inhibiting organophosphate (Paraoxon) on blood pressure and on the electrical activity of preganglionic efferent sympathetic nerves were studied in rats.

It was found, that the organophosphate facilitates the preganglionic sympathetic action potentials, throughout its pressor effect in first stage or depressor effect in late stage on the rat. The apparent dissociation of the stimulating effect on preganglionic action potentials from the depressor effect induced by Paraoxon was abolished in PAM-treated rats. A pretreatment with N-methylatropine and of small doses of atropine (0.5 mg/kg i.v.) also abolished the apparent dissociation.

Toxogonin and large doses of atropine antagonized completely all effects of Paraoxon.

In reserpine-treated rats Paraoxon facilitates the electrical activity of sympathetic preganglionic nerves, whereas the pressor effect was depressed or abolished. Similarly, α-receptor blocking drugs (p.e. phentolamine) depressed or abolished only the pressor effect induced by Paraoxon.

A central cholinergic mechanism for the studied effects is discussed.
