Cytophotometrische und autoradiographische Untersuchungen des Wachstums von Ascitestumoren

Summary

Generation time and duration of the S-phase of a diploid, hyperdiploid and a related hypertetraploid Ehrlich-Ascites-Tumor were determined by autoradiography. The DNA content of the different cells was determined cytophotometrically, and proved the homogenity and the genetic relationship of the tumors. The ratio of the generation times of the diploid tumors to those of the hyper tetraploid was about 1:2. Similar studies of a triploid MC 1 M ascites sarcoma showed, however, that the DNA content did not affect the generation time. The selective growth-advantage of cells of a lower ploidy of a tumor is discussed.

Zusammenfassung

Generationszeit und Dauer der S-Phase eines diploiden, eines hyperdiploiden und des davon abgeleiteten hypertetraploiden Ehrlich-Ascitestumors wurden autoradiographisch ermittelt. Mit cytophotometrischen Messungen des DNS-Gehaltes wurde die Homogenität der untersuchten Tumoren geprüft und eine enge Verwandtschaft der Tumoren bestätigt. Die Generationszeiten der diploiden Tumoren verhalten sich zu der des hypertetraploiden annähernd wie 1:2. Ähnliche Untersuchungen an einem triploiden MC 1 M-Ascitessarkom zeigen jedoch, daß eine generelle Beeinflussung der Generationszeit durch den DNS-Gehalt nicht besteht. Ein selektiver Wachstumsvorteil niedriger-ploider Zellen eines Tumors wird diskutiert.
