Zusammenfassung
Mit einer 0,25%igen Lösung von Methylcholanthren in Benzol wurden weiße Mäuse einmal wöchentlich über längere Zeit gepinselt. Im Stadium der Hyperplasie, der Papillome und der Carcinome wurden Gewebsproben für die autoradiographische Untersuchung nach einmaliger Injektion von 3H-Thymidin entnommen.
In der hyperplastischen Epidermis und im Papillom besteht keine verlängerte Lebensdauer der postmitotischen Zellen gegenüber der normaler mehrschichtiger Epithelien. Dagegen kommt es bei der Cancerisierung der Epidermis über das Stadium der Hyperplasie bis zum Carcinom zu einer kontinuierlichen Vrekürzung der Generationszeit als Ausdruck der verstärkten Zellproliferation. Sie beträgt in der normalen Epidermis rund 150 Std, in der hyperplastischen 56 Std und im Plattenepithelcarcinom der Haut 32 Std. In analoger Weise erhöht sich die Mitoserate. Die DNS-Synthesephase bleibt mit 8 Std annähernd unverändert, während die prämitotische Ruhephase bis zum Carcinom hin eine zunehmende Schwankung aufweist, die jedoch nicht über die Länge der DNS-Synthesephase hinausgeht. Mithin besteht bei der Umwandlung der normalen Epidermiszelle in eine Krebszelle die Hauptveränderung des Wachstums in einer Verkürzung der Differenzierungsphase der sich teilenden Zellen.
Summary
The skin of albino mice was painted weekly with a 0.25% solution of methylcholanthrene in benzol over a prolonged period. After a single injection of H3-thymidine biopsies were taken of the hyperplasia, the papillomata, and the carcinomas for radioautographic studies. In the hyperplastic epidermis and in the papillomata the postmitotic cells did not persist for prolonged periods as did those of normal multilayered epithelia. On the contrary, with the cancerization of the epidermis through the stage of hyperplasia to carcinoma there was a continuous reduction in the generation time, indicating an increased proliferation of cells. In the normal epidermis this time was about 150 hours, in the hyperplastic 56 hours, and in the cutaneous squamous cell carcinoma 32 hours. In an analogous manner the mitotic rate increased. The phase of DNA synthesis remained approximately unchanged at 8 hours, whereas the premitotic resting phase up to the carcinoma showed an increasing fluctuation which, however, did not exceed the length of the phase of DNA synthesis. Thus, with the change of the normal epidermal cell into a cancer cell the main change of growth consists of a shorting of the differentiation phase of the dividing cells.
