Zusammenfassung
Kulturen von solidem Yoshida-Sarkom und Walker-Carcinom der Ratte nehmen bei Zusatz von Serum von Ratten, das 48 Std nach einer partiellen Entfernung der Leber gewonnen wurde, aus dem Nährboden um 27–84% weniger an radioaktivem Phosphat auf als bei Zusatz von Serum normaler Ratten. In vitro suspendierte Zellen aus Ascites-Geschwülsten der gleichen Stämme und Suspensionskulturen von Walker- und HeLa-Carcinom nehmen aus dem Suspensionsmittel bei Zusatz von Serum teilhepatektomierter Ratten ebenfalls signifikant weiniger Phosphat auf als bei Zusatz von normalem Serum. Eine Erklärung für diese Hemmwirkung auf die Phosphorylierung ist nicht im Substratmangel zu finden. Suspensionen von Zellen des Ehrlichschen Mäuse-Ascites-Carcinoms nehmen dagegen in einem Medium, das Serum teilhepatektomierter Tiere enthält, um 10–27% mehr Phosphat auf als unter Zusatz von Serum normaler Ratten. Das gleiche, von teilhepatektomierten Tieren gewonnene Serum wirkt demnach zum gleichen Zeitpunkt organspezifisch und nicht artspezifisch stimulierend auf die Phosphorylierungsprozesse der Leber und spezifisch hemmend auf die Phosphorylierung bestimmter Impftumoren und Tumorkulturen.
Summary
Studies were made of cultures of solid Yoshida sarcoma and Walker carcinoma of the rat. These tumors take up from the culture medium from 27% to 84% less radioactive phosphate after the addition of serum of rats obtained 48 hrs. after a partial hepatectomy than when serum from normal rats is added. Likewise, in-vitro suspended cells from the ascites tumors of the same strains and culture suspensions of the Walker and HeLa carcinoma take up significantly less phosphate from the culture medium when the serum of partially hepatectomized rats is added than when normal serum is used. An explanation of this inhibitory effect on the phosphorylation is not to be found in the substrate. In contrast, suspensions of cells of the Ehrlich mouse ascites carcinoma take up from 10% to 27% more phosphate from the medium containing serum from partially hepatectomized rats than when serum of normal rats is added. Hence, serum from partially hepatectomized animals may act in two ways: it may simultaneously be organ specific but not species specific stimulating the processes of phosphorylation in the liver. On the other hand, it may specifically inhibit the phosphorylation of certain transplanted tumors and tumor cultures.
