Summary
The relations between the size of muscle fibers, motor endplates, and nerve fibers were studied under experimental conditions. In the soleus muscle of the rat, hypertrophy or atrophy of the muscle fibers were induced by elimination of synergist function or by tenotomy of the soleus. After cholinesterase staining of the motor endplates, isolated muscle fibers were used for the appropriate measurements.
In the normal soleus muscle, a statistically significant linear relationship was found between the ChE-positive areas of the motor endplates, the socalled „synaptic areas”, and the volume of the respective muscle fibers. Thus the earlier findings (Anzenbacher und Zenker, 1963) of a size relationship between motor endplate and muscle fiber volume are now confirmed in more precise terms.
However, this linear relationship is lost in the hypertrophic as well as in the atrophic soleus muscle.
In experimental hypertrophy, the increase in muscle fiber volume is accompanied by an increase of the „endplate field” (given by the longitudinal and transverse diameters of the motor endplates) as well as by a less prominent increase of the synaptic areas. The percentage of this increase corresponds with that of the muscle fiber volume.
In experimental atrophy, the decrease in muscle fiber volume is accompanied by a decrease of the endplate field, but without any significant change of the size of the synaptic areas. The ChE-positive subunits of the motor endplates are of the same size and number as in the controls, but more densely packed.
In the hypertrophic muscle, there is also a significant increase of the diameters of the preterminal motor nerve fibers, as compared with untreated controls.
Zusammenfassung
Der M. soleus der Ratte wurde durch Ausschaltung der Synergisten zur Aktivitätshypertrophie bzw. durch Tenotomie zur Inaktivitätsatrophie gebracht und zum Studium der Größenabhägigkeit von Muskelfasern, motorischen Endplatten und Nerven herangezogen. Die Messungen wurden an isolierten Muskelfasern vorgenommen, deren Endplatten durch eine ChE-Reaktion dargestellt worden waren.
Im normalen Muskel ergab sich eine signifikante lineare Beziehung zwischen den ChE-positiven Flächen der motorischen Endplatten, den sog. synaptischen Flächen, und dem Volumen der zugehörigen Muskelfasern. Damit konnte die ursprünglich von Anzenbacher und Zenker (1963) erkannte Größenbeziehung zwischen motorischer Endplatte und Muskelfaser-volumen präzisiert werden.
Sowohl nach Hypertrophie als auch nach Atrophie ging diese lineare Beziehung aber verloren.
Die Muskelfaser-Volumenzunahme bei Hypertrophie ist von einer Vergrößerung des sog. Endplattenfeldes (=gegeben durch Länge und Breite der motorischen Endplatte) als auch von einer etwas geringeren aber prozentuell der Muskelfaservolumenzunahme analogen Vergrößerung der synaptischen Fläche begleitet.
Im Falle der Atrophie kam es zwar zu einer Verkleinerung des Endplattenfeldes, jedoch ohne signifikante Veränderung der synaptischen Fläche, d. h. daß die ChE-positiven Subelemente der Endplatten nur näher aneinanderrückten.
Im hypertrophierten Muskel ließ sich auch eine signifikante Kaliberzunahme im terminalen Bereich der motorischen Nervenfasern feststellen.
