IX. Metabolism of estrone by microsomes from rat liver and transplanted hepatocellular carcinomas including minimal deviation (Morris 5123 C, 7800, 9121) hepatomas

Summary

Estrone metabolizing enzymes of microsomes from normal rat liver, minimal deviation hepatomas (5123 C, 7800 and 9121) and a fast growing transplanted hepatoma (DENA-1) were assayed. The consumption of estrone-[16-14C], the formation of estradiol-(17β) and the formation of protein-bound and watersoluble metabolites was followed.

The fast growing hepatomas had lost the ability to inactivate estrone. The inactivation of estrone by the minimal deviation hepatomas tested was considerably slower as compared with normal liver.

Zusammenfassung

Der Stoffwechsel von Oestron-[16-14C] durch Mikrosomen von normaler Rattenleber, Minimalabweichungshepatomen (5123 C, 7800 und 5121) sowie einem schnell wachsenden Hepatom (DENA-1) wurde untersucht. Gemessen wurden der Substratverbrauch, die Bildung von Oestradiol-(17β) sowie von proteingebundenen und wasserlöslichen Metaboliten.

Während das schnell wachsende Hepatom die Fähigkeit zur Oestroninaktivierung vollständig verloren hat, zeigen die Minimalabweichungshepatome einen gegenüber Normalleber verminderten Oestrogenstoffwechsel.
