Nicotinsäureamid, Nicotinsäure und Thiamin werden durch die carcinostatische Äthylenimin-Verbindung Bayer A 139 alkyliert. Die Reaktion erfolgt bei pH 6 wesentlich rascher als bei pH 7,4. Eine Schutzwirkung gegen die Hemmung der Glykolyse von Ascites-Tumorzellen durch A 139 besitzt von diesen 3 Substanzen lediglich Nicotinsäureamid.

Versuche über das Eindringen von Nicotinsäure und Nicotinsäureamid in Ascites-Tumorzellen zeigen, daß beide Substanzen leicht in das Zellinnere gelangen. Permeabilitätsschranken sind demnach nicht für die Wirkungslosigkeit von Nicotinsäure beim Glykolyseschutz vor A 139 verantwortlich. Es wird eine Methode angegeben, mit der eine Vortäuschung des Eindringens durch Adsorption an den Zelloberflächen ausgeschlossen werden kann.

Die Alkylierung von Nicotinsäureamid durch A 139 erfolgt unter Konzentrationsbedingungen, wie sie im manometrischen Versuch mit Ascites-Tumor-zellen vorliegen, so langsam, daß nur ein geringer Prozentsatz des A 139 durch Alkylierung abreagiert. Daher ist für den Schutz vor der Glykolysehemmung durch A 139 nicht die nichtenzymatische Alkylierungsreaktion, sondern die Beteiligung des Nicotinsäureamids an der DPN-Synthese verantwortlich. Das Ausbleiben eines Schutzes durch Nicotinsäure dürfte auf der Unfähigkeit von Ascites-Tumorzellen beruhen, Nicotinsäure für die DPN-Synthese zu verwenden.

Nicotinamide, nicotinic acid, and thiamine are alkylated by the carcinostatic ethelene imine compound Bayer A 139. The reaction is considerably more rapid at pH 6,0 than it is at pH 7,4. With respect to the inhibition of glycolysis in ascites tumor cells by A 139, of these three substances only nicotinamide possesses a protective effect.

Experiments on the penetration of nicotinic acid and nicotinamide into ascites tumor cells show that the two substances easily penetrate into the inner of the cells. Accordingly, differences in permeability are not responsible for the ineffectiveness of nicotinic acid in protecting glycolysis from A 139. A method is communicated, by means of which a simulation of penetration by adsorption on the cell surfaces can be excluded.

With the conditions of concentrations applied in manometric experiments on ascites tumor cells, the alkylation of nicotinamide by A 139 proceeds so slowly that only a small percentage of A 139 reacts. Therefore not the nonenzymic alkylation but the participation of nicotinamide in DPN synthesis is made responsible for the protection from the inhibition of glycolysis by A 139. The lack of protection by nicotinic acid may be due to the inability of ascites tumor cells to utilize nicotinic acid in DPN synthesis.
