Zusammenfassung
In Fortsetzung der vorher mitgeteilten Untersuchungen werden in den gleichen Basalzellkernen der Portio uteri die Gallocyaninchromalaun- und die Fastgreen-Absorptionen cytophotometrisch bestimmt. Die Gallocyaninchromalaun-Absorption nimmt bereits in den Basalzellen des abnormen Epithels zu, was bei konstantem DNS-Gehalt als eine absolute RNS-Vermehrung zu deuten ist. Im abnormen, unruhigen, atypischen und präinvasiven Epithel verhält sich in den Basalzellen die Gallocyaninchromalaun-Absorption parallel der Feulgen-Absorption, die DNS: RNS-Relation bleibt also konstant. Die Fastgreen-Absorption steigt vom normalen über das abnorme und unruhige zum atypischen Epithel fast linear an. Die relative Fastgreen-Absorption bei Bezug auf die Feulgen-Absorption, auf die Gallocyaninchromalaun-Absorption und auf das Kernvolumen erreicht ihr Maximum in den diploiden Komplexen des präinvasiven Epithels. Hier findet der Umschlag von der reaktiven Epithelveränderung in das Carcinom statt; die nachfolgenden Infiltrationen sind Ausdruck des Wachstums der in den Stadien des präinvasiven Epithels entstandenen Krebszellen. Für die allgemeine Cytogenese des Krebses wird gefolgert, daß in der kleinkernigen Zellform des präinvasiven Epithels eine Neuorientierung der RNS-Eiweiß-Komponenten des Zellkernes eintritt, welche die Carcinogenese charakterisiert. Schließlich werden die erhobenen Befunde auf die Problematik des sog. Oberflächencarcinoms angewandt und die biologische Stellung und Wertung der einzelnen präcancerösen histologischen Veränderungen erörtert.
Summary
In continuation of the preceding studies, the gallocyanin-chromalaun and fast-green absorption of the nuclei of the same basal cells of the portio were determined cytophotometrically. The gallocyanin-chromalaun absorption increased in the basal cells of the abnormal epithelium. With a constant content of DNA, this was interpreted as meaning an absolute increase of RNA. In the abnormal, dysplastic, atypical, and pre-invasive epithelia, the gallocyanin-chromalaun absorption ran parallel with the Feulgen absorption. In other words, the relation of DNA: RNA remained constant. The fast-green absorption increased almost linearly from the normal epithelium through the abnormal and dysplastic to the atypical epithelium. In regards to the Feulgen absorption, the gallocyanin-chromalaun absorption, and to the nuclear volume, the relative absorption of fastgreen reached its maximum in the diploid complexes of the pre-invasive epithelium. Here the transformation of the reactive epithelium into the carcinoma took place. The subsequent invasion is an experssion of the growth of cancer cells developing in the pre-invasive stages. For the general cytogenesis of the cancer it is concluded, that in the small nuclear form of the pre-invasive epithelial cell a new orientation of the RNA-protein component of the nucleus occurs; this characterizes the carcinogenesis. The findings are applied to the problem of the so-called carcinoma in situ. The biological significance and the value of the individual histological precancerous changes are discussed.
