Zusammenfassung
Verschiedene Formen des Portioepithels wurden karyometrisch und cytophotometrisch nach Feulgen-Färbung analysiert: Das normale Portioepithel hat in der Basalis und in der Parabasalis vorwiegend diploide Zellkerne, deren Volumina in der Parabasalis aber doppelt so groß sind wie in der Basalis. Dies wird auf eine funktionelle Kernschwellung in den Parabasalzellen bezogen. Im abnormen Epithel sind bei konstantem DNS-Gehalt auch die Kerne der Basalzellen vergrößert; im unruhigen Epithel treten in der Parabasalis überwiegend tetraploide Kerne auf, die außerdem die Zeichen einer lebhaften DNS-Synthese erkennen lassen. Im atypischen Epithel folgen die Basalzellkerne nach und werden in ihrer Mehrzahl tetraploid. Dies ist das Bild des sog. “Oberflächencarcinoms”. Das “präinvasive” Epithel ist durch kleinkernige Zellen gekennzeichnet, die diploid und nur halb so groß sind wie die Zellkerne des atypischen Epithels. Diese plötzliche Ploidiereduktion wird in Analogie zu experimentellen Befunden an der Rattenleber als charakteristisch für die Carcinogenese angesehen. Die beiden untersuchten Portiocarcinome haben einmal einen hyperdiploiden und einmal einen annähernd tetraploiden DNS-Gehalt pro Zellkern mit breiter Streuung. Für eine progressive Aneuploidie, die kontinuierlich in das Carcinom übergeht, findet sich karyometrisch und cytophotometrisch kein Anhalt, weswegen die Aneuploidie der Tumoren als unspezifische Folge des überstürzten Wachstums angesprochen wird.
Summary
Different type of ectocervical epithelium (portio uteri) were studied karyometrically and cytophotometrically with the Feulgen stain. The normal epithelium of the portio had diploid nuclei in the cells of the basal and parabasal layers. The nuclei in the parabasalis were twice the size of those in the basalis. This was related to a functional nuclear swelling in the parabasal cells. In the abnormal epithelium the nuclei of the basal cells are were enlarged havins a constant content of DNA. In the dysplastic epithelium chiefly tetraploid nuclei appear en the paraz basal layer indicating an active synthesis of DNA. In the atypical epithelium the basal cell nuclei change too becoming predominately tetraploid. This represents the carcinoma in situ. The “preinvasive” epithelium is characterized by cells with small nuclei which are diploid and only half as large as the nuclei of the atypical epithelium. In analogy to experimental findings in the rat liver this sudden reduction of ploidy is looked upon as characteristic of carcinogenesis. The two carcinomas of the portio studied shows both a hyperdiploidic DNA content/nucleus and a DNA content approaching a tetrapolidy with a broad dispersion. There is no karyometric and cytophotometric evidence of a progressive aneuploidy which advanced successively to the carcinoma, as a reason why the aneuploidy of the tumor is considered as a nonspecific result of the uncontrolled growth.
