Zusammenfassung
Elektrophoretische Fraktionierung löslicher Proteine aus Goldhamsterniere im Stärkegel (Veronalpuffer pH 8,6) lieferte wenigstens 24 Banden. In vergleichend untersuchten Extrakten aus einem transplantablen Goldhamsternierencarcinom (Horning) fehlten bei einer schlechteren Auflösung der Banden mindestens 2 kathodisch und 2 anodisch laufende Fraktionen. In zusätzlichen Versuchen zur Lokalisierung bestimmter Fermentaktivitäten auf dem Pherogramm wurden signifikante Unterschiede in der Intensitäten auf dem Pherogramm wurden signifikante Unterschiede in der Intensitätsverteilung der Lactatdehydrogenase-Isoenzyme zwischen Normal- und Tumorgewebe festgestellt. Die nachweisbaren 5 Isoenzyme besitzen in beiden Geweben anodische Beweglichkeit.
Die Zuordnung der Proteinbanden erfolgte mit Hilfe der relativen Wanderungsgeschwindigkeit (RAlb.-Werte) gegen die Albuminbande bzw. auf Grund der elektrophoretischen Beweglichkeit im Stärkegel (u St).
Ultrazentrifugation lieferte bei beiden Gewebeextrakten bis zu 6 Sedimentationsgipfel. Die Fraktionierung war stark von der Eiweißkonzentration abhängig.
Summary
The fractionation of soluble renal proteins of the golden hamster by starch-gel electrophoresis (buffered at pH 8.6 with veronal) gave at least 24 bands. Comparable extracts of a transplantable renal carcinoma (Horning) of the golden hamster gave a poorer separation of the bands; they lacked at least two bands migrating to the anode and two to the cathode. In additional studies of the localization of enzyme activities by the pherogram, significant differences in the intensity of the lactic dehydrogenase isoenzymes were found between the normal tissue and the tumor. The five isoenzyme detected in both tissues migrated towards the anode. The classification of the protein bands was achieved by comparing them with the relative migration velocity of the albumin bands (RAlb.) and by the electrophoretic motility in starch-gel (u St) respectively.
Ultracentrifugation of both tissue extracts disclosed up to 6 sedimentation peaks. The fractionation (number of peaks) was dependent on the concentration of the protein.
