Zusammenfassung
Zellen eines Dauerstammes des Walker-Carcinoms in vitro erhielten Mediumzusätze von normalem Kaninchenserum und von Kaninchenserum nach mehrwöchiger Immunisierung mit Zellen des Walker-Ascites-Carcinoms und mit Rattenblut. Die eintretende Reaktion wurde phasenoptisch beobachtet und mikrokinematographisch registriert. Das Serum einzelner, unbehandelter Kaninchen ist primär cytotoxisch. Diese Reaktion ist komplementunabhängig. Serum von Kaninchen nach Immunisierung mit Rattenblutzellen führt zur reversiblen Abkugelung der 2-dimensional orientierten Zellen. Absättigung dieses aktiven Serums mit normalem Rattenorganbrei hebt seine nachweisbare Wirkung auf.
Zusatz von Kaninchenserum nach Immunisierung mit Walker-Ascites-Carcinom führt zur vollständigen Cytolyse aller Zellen der Kultur. Diese Cytolyse tritt als plötzliche Reaktion nach einer veränderungsfreien Latenzzeit von 10 min auf. Die Reaktion ist komplementabhängig. Die Absättigung des Serums mit Rattenorganbrei hat nur eine Verlängerung der Latenzzeit vor der Cytolyse zur Folge.
Die sichtbare Immunreaktion wird an Hand des mit Zeitraffung gefilmten Ablaufes geschildert.
Summary
Cells of a permanent strain of Walker carcinoma grown in vitro were treated separately with normal rabbit serum, with serum from rabbits immunized for several weeks with cells of Walker ascites-carcinoma, or with rat blood. The reaction was studied with phase microscopy and photographed with microcinematography. The serum of untreated rabbits was cytotoxic, the reaction being independent of complement. With serum of rabbits immunized with rat blood, the flattened cells became globular, but this change was reversible.
Neutralization of the serum with homogenized normal rat organs abolished the effect. Serum of rabbits immunized with Walker ascites-carcinoma caused complete cytolysis of all cells of the culture. This cytolysis, preceded by no changes, appeared suddenly after a latent period of 10 min after adding serum. This reaction requires complement. Neutralization of the serum with homogenized rat organs caused only an increase in the latent period. The sequence of events in the visible immune reaction are depicted in photographs obtained by time-lapse microcinematography.
