Zusammenfassung
Nach Zusatz von 2-Desoxyglucose (DOG) zu einer Pyruvat enthaltenden Suspenpension von Ehrlich-Ascitestumorzellen der Maus kommt es zu einem beträchtlichen Anstau von Citrat. Diese Wirkung der 2-Desoxyglucose ist auch unter den Bedingungen des endogenen Stoffwechsels (Suspensionsmittel: substratfreie Salzlösung) feststellbar, jedoch weniger ausgeprägt und von kürzerer Dauer.
Die Umsatzrate des Pyruvats wird durch DOG erniedrigt. Eine Lactatbildung findet nicht statt.
Pyruvat bewirkt einen Anstau von α-Ketoglutarat, diese Bildung von Ketoglutarat wird durch DOG beträchtlich gehemmt.
Die von früheren Autoren als „Crabtree-Effekt” gedeutete Verminderung des Sauerstoffverbrauches der endogenes Substrat veratmenden Zellen nach Zusatz von DOG findet eine zwanglose Erklärung in der beobachteten Hemmung der Citratverwertung.
Summary
After adding 2-deoxyglucose (DOG) to a suspension of Ehrlich ascites tumor cells of the mouse containing pyruvate, a considerable amount of citrate accumulates. This effect of the 2-deoxyglucose is detectable also under conditions of endogenous metabolism (suspending medium: salt solution free from substrate); however, it is less pronounced and of shorter duration.
The turnover rate of the pyruvate is reduced by the DOG. Lactate is not formed. The pyruvate induces an accumulation of α-ketoglutarate. This formation of ketoglutarate is inhibited considerably by the DOG. The reduction of the O2 consumption of the cells consuming endogenous substrate which occurs after the addition of DOG, interpreted by other authors as „Crabtree effect”, can best be explained by the observed inhibition of the utilization of citrate.
