Untersuchungen zur Ultrastruktur von Keratinomyces Ajelloi Vanbreuseghem 1952

Summary

The ultrastructure of aerial hyphae and macroconidia of the dermatophyte Keratinomyces Ajelloi is described. Cell walls and septa were composed of several distinct layers. Septa occurred in different stages of their formation. The occurrence of Woronin bodies (septal pore plugs) indicates the presence of septal pores. The cytoplasma was surrounded by an electron-dense cell membrane which was partially interrupted by invaginations and transformed into small curved fragments or vesicles. Lomasomes were reported for the first time in dermatophytes. Within the cytoplasma mitochondria, tubular or lamellar structures which are designated as endoplasmic lamellae, and nuclei without nucleoli could be demonstrated. electron-dense inclusions (probably polysaccharide storage granules) were accumulated mainly in the hyphae. In addition, electron transparent unidentified material was found in the macroconidia. Further cell constituts were diffusely dispersed cytoplasmic granula (ribosomes) and, within the cells of macroconidia, large vacuoles.

Zusammenfassung

Die Ultrastruktur von Luftmycel und Makroconidien des Dermatophyten Keratinomyces Ajelloi wurde beschrieben. Zellwände und Septen wiesen einen schichtartigen Bau auf. Die Septen konnten in verschiedenen Entwicklungsstadien dargestellt werden. Der Nachweis von Woronin-Körperchen (“septal pore plugs”) deutet auf das Vorhandensein von Septumporen hin. Das Cytoplasma ist von einer elektronendichten Zellmembran umschlossen, die teilweise Invaginationen aufweist und an einigen Stellen in gebogene Bruchstücke sowie kleine Vesikel umgewandelt ist. Lomasomen wurden erstmals bei einem Dermatophyten beschrieben. Im Cytoplasma finden sich Mitochondrien, tubuläre oder lamellenartige Strukturen, die als endoplasmatische Lamellen zu bezeichnen sind, sowie Zellkerne ohne Nucleolus. Als Zelleinschlüsse stellten sich grobschollige, elektronendichte Ansammlungen (wahrscheinlich gespeicherte Polysaccharide) vor allem in den Hyphen dar, während in den Makroconidien zusätzlich ein elektronendurchlässigeres, nicht identifiziertes Material gefunden wurde. Als weitere Zellbestandteile fanden sich diffus im Cytoplasma verteilte, feinkörnige Granula (Ribosomen) sowie große Vacuolen in den Makroconidien-Kammern.
