Summary
The electron microscopic study of Darier's disease shows a structural alteration of the peripheral cytoplasm, where the tonofilaments are separated from the desmosomes. The number of desmosomes is reduced. Because the cohesion between the cells is diminished, they separate from each other. The nucleo-cytoplasmic ratio is modified in favour of the nucleus. Keratohyalin granules are already formed in the lowest layers of the epidermis. The “grain” results from a process of keratinisation, where the cytoplasm becomes amorphous and looses his normal filamentous structure. The process can be accompanied by pycnosis of the nucleus. In the stratum granulosum intracytoplasmic masses are described, the meaning of which is not yet fully understood.
Zusammenfassung
Die Ultrastruktur der Darierschen Krankheit zeigt eine strukturelle Änderung des peripheren Zellcytoplasmas, wobei die Tonofilamente von den Desmosomen getrennt sind; die Desmosomen werden weniger zahlreich. Wegen mangelnder Kohäsion trennen sich die Zellen nachher voneinander ab. Die nucleo-cytoplasmatische Relation wird zugunsten des Kernes verschoben. Keratohylingranula bilden sich schon in den tiefsten Schichten der Epidermis. Ein „grain” entsteht durch einen Keratinisationsprozeß, wobei das Cytoplasma amorph wird und seine normale filamentöse Struktur verliert; der Prozeß kann mit einem Kernzerfall begleitet sein. Im Stratum granulosum wird eine intracytoplasmatische Masse beschrieben, deren Bedeutung noch ungeklärt ist.
