Summary
Out of 17 strains of 14 different Bacillus species, Bacillus cereus ATCC 7064 produced the highest levels of p-nitrophenyl-α-D-glucopyranoside-hydrolyzing α-glucosidase. Maximal enzyme yield was achieved after 6 h of cultivation, at an initial pH of 6.5–7.0, on a medium containing 2% soluble starch, 2% peptone, 0.3% yeast extract, 0.05% meat extract, 0.3% K2HPO4 and 0.1% KH2PO4. When grown on 2% starch, B. cereus synthesized α-glucosidase in the cytoplasm. At a starch concentration of less than 1%, however, the enzyme appeared in the culture broth. This accumulation was maximal at 0.4% starch when the extracellular enzyme amounted to 54% of the total enzyme produced.
