Summary
A method for the estimation of nicotine in blood and tissues by gas liquid chromatography is described. By this method nicotine can be estimated in concentrations of 0.01 μg/ml blood resp. g tissue by using ca. 5 g of material. The use of greater quantities of material increases the sensitivity. Nicotine may also be separated from its metabolites by this method. Besides description of the method statements with regard to precision, accuracy and specificity are made. In venous blood from smokers nicotine has been estimated 10 min after smoking one cigarette ranging from 0.005 to 0.064 μg/ml blood.
Zusammenfassung
Mit der beschriebenen Methode kann Nicotin bis zu einer Konzentration von 0,01 μg/ml Blut bzw. g Gewebe bei Einsatz von ca. 5 g Material quantitativ bestimmt werden. Bei Einsatz größerer Gewebemengen kann die Empfindlichkeit gesteigert werden. Nicotin kann mit dieser Methode auch neben seinen Metaboliten identifiziert werden. Außer der Beschreibung der Methode werden Angaben zur Genauigkeit, Richtigkeit und Spezifität gemacht. Bei Rauchern werden 10 min nach dem Rauchen einer Zigarette 0,005–0,064 μg Nicotin/ml im Venenblut gefunden.
