The protective effect of acetylsalicylic acid on laser-induced venous thrombosis in the rat

Summary

The protective effect of acetylsalicylic acid (ASA) against thrombosis was investigated in the jugular vein of rats at sites of endothelial trauma induced with a high energy output ruby laser. To examine quantitatively thrombus formation and the effects of ASA, the thrombi were dissected out of the jugular vein 4 h after the laser-induced injury and weighed accurately. The results obtained with this standardized and quantitative technique indicate that ASA has a significant protective effect against laser-induced thrombosis. Inhibition of thrombus growth occurred when ASA was orally administered in doses of 3 and 10 mg/kg simultaneously with the laser stimulus. Increasing the ASA to 30 and 100 mg/kg did not produce further inhibition. Administration of ASA 16 h before induction of thrombosis also markedly inhibited thrombus formation. ASA probably acts to inhibit thrombus formation by inhibiting platelet aggregation. Administration of ASA 24 h after induction of thrombosis did not significantly alter the size or weight of the thrombi, indicating that ASA is not able to produce thrombolysis.
