Effects of ultraviolet light on human dermal collagen in vitro

Summary

Under the influence of ultraviolet light, changes in hydroxyproline content and changes in viscosity of neutral salt soluble and of acid soluble collagen of human dermis were investigated. With increasing irradiation time, an augmentation of hydroxyproline content has been demonstrated. This observation which applies equally for neutral salt soluble and for acid soluble collagen proves a hydroxylation of proline by ultraviolet light. Hydroxyproline formation might be effectuated either directly (via oxygen) or indirectly (via hydrogen peroxide). — With increasing irradiation time, viscosity of neutral salt soluble and acid soluble collagen of human dermis decreases. This effect is explained with a degradation of primary collagen chains under the influence of ultraviolet light.

Zusammenfassung

Die vorliegenden Untersuchungen beschäftigen sich mit dem Einfluß von Ultraviolettlicht auf Kollagen der menschlichen Dermis in vitro. Neutralsalzlösliches und säurelösliches Kollagen wurden bestrahlt; nach verschieden langer Bestrahlungsdauer wurden der Hydroxyprolingehalt und die Viscosität bestimmt. Mit zunehmender Bestrahlungsdauer ergab sich in beiden Kollagenlösungen ein Anstieg des Hydroxyprolingehaltes und eine Abnahme der Viscosität. Die Zunahme des Hydroxyprolins geht auf eine Hydroxylierung von Prolin unter der Einwirkung von Ultraviolettbestrahlungen zurück; die Hydroxylierung erfolgt entweder direkt oder indirekt (über Bildung von Wasserstoffperoxyd). Die Abnahme der Viscosität ist durch Veränderungen der Kollagenstruktur (Lösung der Primärketten) als Folge der Ultraviolettbestrahlungen bedingt.
