Zur quantitativen Bestimmung von Thebain in Papaver bracteatum

Processes of drying, homogenisation and extraction of Papaver bracteatum are investigated with respect to a quantitative analysis of thebaine. Based on these results a method is proposed by which the fresh plant material is lyophilised, homogenised by ball milling and extracted three times with 5% acetic acid. The pure extract of thebaine is then obtained by means of an Extrelut column (Merck). By this extraction scheme followed by quantitative HPTLC the relative standard deviation of thebaine content in Papaver bracteatum is 2.3%.

Zusammenfassung

Es werden Verfahren der Trocknung, Zerkleinerung und Extraktion von Papaver bracteatum in Hinblick auf eine quantitative Thebainbestimmung überprüft. Auf Grund der Ergebnisse wird eine Methode vorgeschlagen, nach der die Droge gefriergetrocknet, anschlie\end in einer Kugelmühle zerkleinert, mit 5% iger EssigsÄure in 3 Stufen extrahiert und daraus der Thebainextrakt mit Hilfe einer Extrelut-SÄule (Merck) gewonnen wird. Nach diesem Extraktionsschema mit der anschlie\enden HPTLC-Auswertung betrÄgt die relative Standardabweichung des Thebaingehaltes aus Papaver bracteatum 2,3%.
