Summary
Staining of human rib cartilage by alcian blue at different MgCl2-concentrations shows the well known age-related pattern of acid mucopolysaccharides.
Kryostate sections are completely and intensively coloured already with electrolyte at concentrations of 0.01–0.05 m. Also with higher concentrations all structures are distinctly visible. After fixation, however, the tissue reaches only its full staining intensity following application of medium concentrations, while at higher concentrations the structures become unclear. Mucopolysaccharides which remain unstained using their critical electrolyte concentrations with alcian blue can be demonstrated by acridin organe. Staining intensity however is dependent on age rather than on fixation. Cartilage of new-born babies stained with 0.4 m MgCl2 becomes blue without taking up yellow dye, while with a concentration of 0.9 m MgCl2 on the contrary it is yellow without taking up blue dye. This is due to the high concentration of acid mucopolysaccharides containing only chondroitine sulfates. Alcian blue was not displaced by acridin orange in our experimental conditions. In old rib cartilage yellow colour means absence of alcianophilia, green means partial and blue distinct alcianophilia. By this method even small changes in staining intensity with alcian blue at the critical electrolyte concentrations are demonstrable.
Zusammenfassung
Durch Alcianblaufärbung bei verschiedenen MgCl2-Konzentrationen wird in menschlichem Rippenknorpel die bekannte, alternsbedingte Verteilung der sauren Mucopolysaccharide dargestellt.
Kryostatschnitte zeigen schon bei Elektrolytkonzentrationen von 0,01–0,05 m eine vollständige, intensive Anfärbung. Auch bei hohen Konzentrationen sind noch alle Strukturen deutlich gefärbt. Dagegen erreicht fixiertes Gewebe erst bei mittleren Konzentrationen seine volle Farbintensität, während hohe eine unvollständige Darstellung der Strukturen ergeben.
Die bei den jeweils kritischen Elektrolytkonzentrationen nicht mehr mit Alcianblau anfärbbaren sauren Mucopolysaccharide werden durch Gegenfärbung mit Acridinorange erkennbar. Dabei ist die Färbung mehr vom Alter als von der Fixierung abhängig. Knorpel vom Säugling färbt sich bei 0,4 m MgCl2 blau ohne Aufnahme von gelbem Farbstoff, bei 0,9 m MgCl2 umgekehrt gelb an. Dies entspricht dem hohen Gehalt an sauren Mucopolysacchariden bei ausschließlicher Zusammensetzung aus Chondroitinsulfaten. Eine Verdrängung von Alcianblau bei der Gegenfärbung findet unter den gewählten Bedingungen nicht statt. Im alten Rippenknorpel sieht man bei fehlender Alcianophilie Gelbfärbung, bei teilweiser Alcianophilie Grünfärbung und bei ausgeprägter Alcianophilie Blaufärbung der Strukturen. Auch kleinere Veränderungen der Anfärbbarkeit bei den jeweils kritischen Elektrolytkonzentrationen lassen sich so nachweisen.
