Summary
The effect of hydroxyurea on glycogen, lipoids, and enzymes of liver, kidney, and pancreas is investigated in male Wistar rats by means of histochemistry and electron microscopy. After 4 or more days of hydroxyurea application glycogen and lipoids decrease in the liver, whereas acid phosphatase and acid 5-nucleotidase activity markedly increase. Electron microscopically numerous autophagic vacuoles are visible. The activity of G-6-PDH and cytochrome-oxidase show little increase in the liver. Alkaline phosphatase, LDH, and SDH reactions don't change after hydroxyurea. In opposition to the liver the activities of the acid hydrolases in pancreas remain low as in control animals and decrease in the proximal tubule of the kidney.
A single injection of fluorouracil (50 mg/animal) causes the loss of liver glycogen and an increase of acid phosphatase- and acid 5-nucleotidase-containing lysosomes in the three investigated organs. At the 3rd day after the fluorouracil application the acid hydrolases reach their highest level, whereas the unspecific esterase fall to its lowest activity. The other investigated enzymes show no significant changes after fluorouracil treatment.
Zusammenfassung
An erwachsenen männlichen Wistarratten wird die Wirkung von Hydroxyharnstoff und Fluoruracil auf Reservestoffe, Enzyme und Ultrastruktur von Leber, Niere und Pankreas histochemisch und elektronenmikroskopisch untersucht. Nach 4- und mehrtägiger Behandlung mit Hydroxyharnstoff (100 mg/100 g Körpergewicht) nehmen Glykogen und Lipoide in der Leber ab, während die saure Phosphatase und die saure 5-Nukleotidase deutlich an Aktivität gewinnen. Elektronenmikroskopisch sind vermehrt autophagische Vakuolen sichtbar. Die Aktivitäten der Glukose-6-Phosphatdehydrogenase und der Cytochromoxidase nehmen in der Leber mäßig zu; alkalische Phosphatase, LDH und SDH verändern ihre Aktivität unter der Behandlung nicht. Im Gegensatz zur Leber bleiben die Aktivitäten der sauren Hydrolasen im Pankreas so gering wie bei den Kontrolltieren und nehmen im proximalen Tubulusabschnitt der Niere ab.
Fluoruracil bewirkt nach einmaliger Injektion (50 mg/Tier) in der Leber bereits nach einem Tag einen weitgehenden Verlust des Glykogens und eine Vermehrung der sauren Phosphatase- und sauren 5-Nukleotidase-positiven Lysosomen in allen drei untersuchten Organen. Am 3. Tag nach der Applikation erreichen die sauren Hydrolasen ihre höchste Aktivität, während gleichzeitig die unspezifische Esterase ihr niedrigstes Niveau hat. Die übrigen untersuchten Enzyme zeigen keine deutliche Veränderung unter der Fluoruracilbehandlung.
