Zusammenfassung
Eine Methode zur Analyse von Pentachlorphenol (PCP) in humanem Blutserum bzw. Urin wird beschrieben. Dazu werden die Proben nach Ansäuern des Serums bzw. saurer Hydrolyse des Urins mit Toluol extrahiert und die organische Phase dann mit einer wäßrigmethanolischen Lösung von Triethylsulfoniumhydroxid (TESH) reextrahiert. Die wäßrige Phase wird in den Gas-Chromatographen injiziert, dabei werden saure Substanzen wie PCP pyrolytisch ethyliert. Unter Verwendung von Capillarsäulen und Elektroneneinfangdetektion beträgt die Nachweisgrenze 2 μg/l für Urin and 3 μg/l für Serum. Die Richtigkeit der Methode wurde durch Gaschromatographie-Massenspektrometrie überprüft. Serum- und Urin-Konzentrationen von Personen, die keinen besonderen Kontakt mit PCP hatten und von Personen, die vor einigen Jahren PCP-haltige Holzschutzmittel angewandt hatten, werden berichtet.
Summary
A method to analyze pentachlorophenol (PCP) in human serum and urine is described. Sample preparation involves extraction of PCP with toluene after acidification of serum (0.5 ml) or acidic hydrolysis of urine (1 ml). The toluene extracts are then reextracted by an aqueous-methanolic solution of triethylsulfonium hydroxide (TESH). This phase is injected into the gas chromatograph where a pyrolytic ethylation of acidic compounds like PCP occurs. Using capillary columns for chromatography and EC detection the detection limit is 2 μg/l for urine and 3 μg/l for serum. The accuracy of the method was ensured by gas chromatography-mass spectrometry. Serum and urine concentrations of persons not exposed to PCP as well as of persons, that used wood preserving agents some years ago, are reported.
