Zusammenfassung
Verschiedene Amine wurden mit Natrium-9,10-dimethoxyanthracen-2-sulfonat zu Ionenpaaren umgesetzt, die in eine organische Phase extrahiert und fluorimetrisch bestimmt wurden. pH-Abhängigkeit der Ionenpaarbildung, sowie Einfluß des Extraktionsmittels und der Reagenskonzentration wurden untersucht und ein Fließschema für ein automatisches Verfahren ausgearbeitet. Linearität ergab sich im Bereich von 10 ng/ml bis 2 μg/ml, die Reproduzierbarkeit war besser als 1%, Verschleppungseffekte waren vernachlässigbar und Störungen durch Hilfsstoffe traten nicht auf.
Summary
Several amines were converted with sodium 9,10-dimethoxyanthracene-2-sulphonate to ion pairs, which were extracted from the aqueous phase into an organic solvent and determined by fluorescence. The pH-dependence of the ionpairing, the extraction solvent, and the reagent/amine concentration ratio were investigated using several drug substances with the purpose of establishing an analytical procedure for general use. On the basis of these results a flow-scheme for an automated procedure was developed. The linearity of this system was within the range of 10 ng/ml to 2 μg/ml, reproducibility was better than 1%, the carry-over effect was negligible, and there was no interference by excipients.
