Exploration de l'immunite humorale dans l'Aspergillose: Intérêt de l'E.L.I.E.D.A. (Enzyme linked immuno-electro-diffusion assay)

Abstract

In order to increase the sensitivity of the method of immunoelectrodiffusion (I.E.D.) and to isolate the class of immunoglobulins implicated or involved in the Aspergillose antigen-antibody reaction, we supplemented the I.E.D. by combining or incorporating it with an enzymatic technique. By this method the immune complexes precipitated are taken up by antibodies conjugated to one enzyme and specific to the immunoglobulins of each class. This constitutes the technique of E.L.I.E.D.A. (enzyme-linked-immuno-electro-diffusion assay). The sensitivity and the specificity of the proposed method would allow us to follow the qualitative evolution of the aspergillar antibodies and would lend support to further arguments on the evolutionary character of the inflammatory response.

Résumé

Dans le but de sensibiliser la méthode d'immuno-éléctrodiffusion (IED) et de définir la classe des immunoglobulines impliquées dans la réaction antigène — anticorps de l'Aspergillose, nous avons complété l'IED en la couplant à une technique enzymatique. Pour cela, les immuncomplexes précipités sont traités par des anticorps conjugués à une enzyme et spécifiques des immunoglobulines de chaque classe. Cette association réalise la technique d'E.L.I.E.D.A. (enzyme-linked-immuno-electro-diffusionassay). La sensibilité et la spécificité de la méthode qui s'avèrent remarquables devraient permettre de suivre l'évolution qualitative des anticorps aspergillaires et d'apporter des arguments sur le caractère évolutif de l'affection.
