Abstract

The haptoglobins (Hp) of control and gamma-irradiated Spanish goats were examined. Polyacrylamide gel electrophoresis and Sephadex G-200 chromatography of sera demonstrated that unirradiated controls were ahaptoglobinemic, while irradiated animals developed high Hp levels. An electrophoretic comparison with human Hp phenotypes revealed that goat Hp more closely resembled human Hp 2 than the other two human phenotypes. No electrophoretic band comparable to human Hp 1 appeared in the goat electrophoretic pattern. When goat Hp was tested by immunodiffusion with antiserum to human Hp, no reactivity was observed; however, goat Hp and the three human Hps bound goat, human, and chicken hemoglobins with equal specificity, indicating some degree of structural homology between human and goat Hp. Suggestions as to the effects of irradiation to augment Hp synthesis and speculations about the chain composition and molecular weights of goat Hp are presented.
