Abstract
The influence of GSSG on the activities of all glycolytic enzymes as well as on the activities of glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and glutathione reductase was studied in hemolysates of red cells from newborns and adults.
Of the enzymes studied hexokinase, 6-phosphofructokinase, glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase are inhibited by GSSG considerably.
Glucose-6-phosphate dehydrogenase is less inhibited in hemolysates from red cells of newborns than in those of adults. All other enzymes show no differences.
The inhibition of glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase can be prevented to a large extent by NADP and that of phosphofructokinase by fructose-6-phosphate.
Elevated activity of glutathione reductase in hemolysates produced by the addition of purified enzyme increases the inhibitory action of GSSG.
The content of GSSG is negligible in the red cells of newborns and adults whereas red cells of newborns contain nearly twice the amount of GSH than red cells of adults.
Zusammenfassung
Es wurde die Wirkung von GSSG auf die Aktivitäten sämtlicher Glykolyseenzyme, der Glucose-6-phosphatdehydrogenase, der 6-Phosphogluconatdehydrogenase und der Glutathionreduktase in Hämolysaten aus Erwachsenen-und Neugeborenen-Erythrocyten geprüft. Von den untersuchten Enzymen werden Hexokinase, 6-Phosphofructokinase, Glucose-6-phosphatdehydrogenase und 6-Phosphogluconatdehydrogenase durch GSSG beträchtlich gehemmt.
Die Glucose-6-phosphatdehydrogenase wird in Hämolysaten aus Neugeborenen-Erythrocyten weniger stark durch GSSG gehemmt als in Hämolysaten aus Erwachsenen-Erythrocyten. Die übrigen Enzyme zeigen keine Unterschiede.
Die Hemmung der Glucose-6-phosphatdehydrogenase und der 6-Phosphogluconatdehydrogenase läßt sich durch NADP und die Hemmung der Phosphofructokinase durch Fructose-6-phosphat weitgehend aufheben.
Erhöhung der Glutathionreduktase-Aktivität im Hämolysat durch Zugabe von gereinigtem Enzym verstärkt die GSSG-Wirkung erheblich.
Neugeborenen-Erythrocyten und auch Erwachsenen-Erythrocyten enthalten praktisch kein GSSG, während der GSH-Gehalt in Neugeborenen-Erythrocyten fast doppelt so hoch ist wie in Erwachsenen-Erythrocyten.
