Zusammenfassung
Schon Osborne, der Pionier auf dem Gebiete der Erforschung pflanzlicher Proteine, hat festgestellt, daß die Anwesenheit anorganischer Salze die Wasserlöslichkeit der Prolamine vermindert. Es ist daher ganz unzweckmäßig, Antigenlösungen so herzustellen, daß man Suspensionen von Gliadin in Kochsalz-Phosphat-Lösungen zentrifugiert und den Überstand verwendet. Die Titer der KB-Reaktion von echten Lösungen des Gliadins in alkalisiertem Wasser sind weit höher, der Prozentsatz positiver Reaktionen bei dem Pr.-Test ist größer. Pseudopositive Reaktionen kamen mit dieser Probe nicht vor. Der Gebrauch der Fraktion III zum Antikörpernachweis mittels Pr oder HA ist nicht zu empfehlen, da die Prozentzahlen positiver Reaktionen bei Sprue und Cöliakie zu niedrig sind. Es ist unklar, wie häufig Posivität der Kontrollen vorkommt. Fraktion III ist nicht eine chemisch einheitliche Substanz, wie behauptet wurde1, sondern ein Stoffgemisch. Gliadin setzt sich nach Waldschmidt-Leitz u. P. Metzner 11 aus sieben Komponenten zusammen. Weizengliadin und Roggengliadin sind chemisch nicht identisch11. Es ist anzunehmen, daß nicht alle Prolaminkomponenten bei der Immunisierung gleich wirksam sind. Es könnten Unterschiede zwischen den einzelnen Komponenten bestehen.
Summary
It was found already by Osborne, the pioneer in vegetable protein research, that the presence of inorganic salts reduces the water solubility of prolamines. Hence, it is inexpedient to prepare antigen solutions by centrifuging gliadin suspensions in sodium chloride—phosphate solution and utilizing the supernatant obtained. When using dilute alkali solutions, the titres of such solutions of gliadin are far higher with the complement fixation test and, in celiac disease, the percentage of positiv reactions obtained in the gel diffusion test ist greater. We never had with this test positive reactions in the controls. The use of fraction III—prepared according Frazer—in antibody determinations by hemagglutination or gel diffusion is not advisable, as the number of positive reactions obtained in sprue and celiac disease are too low. Fraction III is not a homogeneous substance. Gliadin consists of seven components (Waldschmidt-Leitz and Metzner 11). Wheat gliadin and rye gliadin are not identical. It may be that the different components are not equally active in immunization.
