The ineffectiveness of desipramine pretreatment on behavioral effects of 6-hydroxydopamine in nialamide-pretreated rats

Abstract

Non-anesthetized rats, pretreated with nialamide (140 mg/kg, 16–18 h earlier) were injected with 6-hydroxydopamine (HBr, 2×250 Μg) into both lateral brain ventricles. Some rats received desipramine (25 mg/kg) 1 h before 6-hydroxydopamine. 6-Hydroxydopamine increased the locomotor activity and produced behavioral stimulation lasting for several hours. On the following days the open-field performance of the rats receiving 6-hydroxydopamine was markedly depressed. Approx. one half of the rats receiving 6-hydroxydopamine died between day 3 and day 10 after the injection. Desipramine pretreatment did not affect the behavioral changes or mortality produced by 6-hydroxydopamine, although it counteracted the depletion of brain noradrenaline brought about by 6-hydroxydopamine. It is concluded that the behavioral changes observed after 6-hydroxydopamine injection in nialamide-pretreated rats are related to the action of 6-hydroxydopamine on dopamine, and not on noradrenaline neurons.
