Résumé
Cette note décrit l'application de la technique de culture en dialyse continue à Nitrobacter. La culture ne peut pas être maintenue en phase exponentielle beaucoup plus longtemps par cette méthode que dans le cas où les nitrates peuvent s'accumuler dans le milieu de culture. Toutefois, la croissance continue à vitesse constante au lieu de s'arrêter complètement, et l'on peut atteindre une concentration de quelque 500 mg de matière sèche au litre. Il est montré que c'est la diffusion du nitrite à travers la membrane qui limite la croissance bactérienne et que si l'on augmentait la quantité de nitrite diffusée une nouvelle limitation de la croissance due cette fois à un apport d'oxygène insuffisant ne tarderait pas à se manifester.
Summary
This note describes the use of continuous dialysis for growing Nitrobacter.
It is not possible to keep Nitrobacter growing exponentially during a much longer time by this method than when nitrate is not dialyzed from the culture medium. However, growth continues at a constant velocity instead of stopping and a cell concentration of about 500 mg per 1 (dry weight) is easily reached. It is shown that nitrite diffusion through the membrane is rate limiting for growth and that increase of the amount of nitrite diffusing through the membrane would soon lead to another rate limiting process, namely oxygen diffusion through the medium from the air bubbles.
