Summary
Gelfiltration offers newpossibilities for studying labelled zinc fixation on plasma proteins. This heavy metal has a great affinity to protein, but its binding is reversible under the effect of pH and chelating substances (Versenate, Penicillamine). — The existence of endogenous chelating substances as well as the chelating effect of Alloxan are not confirmed. — Owing to an appropriate dose of Versenate, it is possible to obtain a chelating effect of 50% of labelled zinc, which defines a zinc fixation index on plasma proteins. Comparing plasma of normal and diabetic subjects decompensated and not yet treated, it is possible to show evidence of a higher fixation in diabetics. This higher fixation capacity is probably the result of an increase in globulin fractions fixing zinc strongly, but it is not possible to assess whether it is a primary or a secondary phenomenon to diabetes. — It is known that proteinuria appears very early, even contemporary to the onset of diabetes and its importance seems quite sufficient to explain zincuria which is increased in diabetics badly decompensated.

La filtration sur gel de dextran offre de nouvelles possibilités d'étudier la fixation du zinc marqué sur les protéines plasmatiques. Ce métal lourd a une très grande affinité pour les protéines, mais sa liaison est réversible sous l'effet du pH acide ou des chélateurs (Versenate ou Pénicillamine). — L'existence de chélateurs endogènes ainsi qu'un effet chélateur de l'Alloxane peuvent être infirmés. — Grâce à une dose appropriée de Versenate, il est possible d'obtenir in vitro une chélation de 50% de zinc marqué, définissant ainsi un index 2 de fixation du zinc sur les protéines plasmatiques. En comparant le plasma de sujets normaux et de sujets diabétiques décompensés et non encore traités, il est possible de mettre en évidence un pouvoir de fixation plus élevé chez les diabétiques. — Cette capacité de fixation plus élevée paraît être liée à une augmentation des fractions protéiques (globulines) qui fixent le zinc fortement, sans qu'il soit possible de dire si elle est un phénomène primaire ou secondaire au diabète. — On sait que la protéinurie est très précoce, même contemporaine de l'apparition d'un diabète et son importance paraît suffisante pour expliquer la zincurie augmentée chez les diabétiques gravement décompensés.
Zusammenfassung
Die Filtration über Dextran-Gel eröffnet neue Möglichkeiten, um die Bindung von markiertem Zink an die Plasmaproteine zu erforschen. Dieses Schwermetall hat eine große Affinität zu Proteinen, seine Bindung daran ist jedoch unter dem Einfluß von saurem Ph und Chelatbildnern wie Titriplex und Penicillamin reversibel. Die Theorien über die Existenz endogener Chelatbildner ebenso wie die über chelatbildende Wirkungen des Alloxans sind nicht mehr aufrechtzuerhalten. — Mit einer geeigneten Dosis von Titriplex läßt sich in vitro eine Chelatbildung mit 50% des markierten Zinks erreichen und damit ein Index für das Fixationsvermögen von Zink an Plasmaeiweißkörper gewinnen. Beim Vergleich der Plasmen von Kontrollpersonen und von unbehandelten, dekompensierten Diabetikern ließ sich bei den Diabetikern ein erhöhtes Bindungsvermögen nachweisen. — Dieses erhöhte Bindungsvermögen scheint uns durch eine Vermehrung der Serumeiweißanteile mit einer ausgeprägten Bindungskapazität für Zink (Globuline) bedingt zu sein, ohne daß man sagen kann, ob es sich dabei um ein primäres oder sekundäres Phänomen beim Diabetes handelt. — Es ist bekannt, daß die Proteinurie sehr frühzeitig, ja oft schon bei der Manifestation des Diabetes auftritt, und ihr Ausmaß scheint uns zu genügen, um die vermehrte Zinkausscheidung beim schwer dekompensierten Diabetiker zu erklären.
