Zusammenfassung
Die Aminosäurezusammensetzung des menschlichen Linseneiweißes zeigt in Rinde und Kern mit zunehmendem Alter signifikante Verschiebungen. In der Rinde junger Linsen liegen die Werte für die glykoplastischen Aminosäuren Glycin, Alanin, Valin, Asparaginsäure, Glutaminsäure und Arginin in signifikant höherer Konzentration vor als in den entsprechenden Kerngebieten. Ähnlich verhalten sich die aromatischen Aminosäuren Tyrosin und Phenylalanin. Mit steigendem Alter sinken die Werte dieser Aminosäuren im Kerngebiet ab. Umgekehrt verhalten sich Leucin, Isoleucin und Lysin.
Die Tatsache, daß der Eiweißstickstoffgehalt der Linse trotz ihres ständigen Wachstums nicht ansteigt und der Nachweis proteolytischer Fermente in skierotischen und kataraktösen Linsen berechtigt zu der Annahme, daß es in den zentralen Linsenbezirken zur Aufspaltung von Protein kommen kann. Die dabei entstehenden Restmoleküle dürften die „pathologische Albuminoidbildung“ sowie die Verschiebung der Aminosäurewerte erklären können.
Summary
The amino acid composition of the human lens protein shows significant changes in both cortex and nucleus with increasing age. In the cortex of young lenses, the glycoplastic amino acids glycine, alanine, asparagic acid, glutamic acid and arginine are present in significantly higher concentration than in the corresponding areas of the lens nucleus. The aromatic amino acids tyrosine and phenylalanine reveal a similar behaviour. With increasing age, the values of these amino acids decrease in the area of the nucleus. Leucine, isoleucine and lysine show the reverse behaviour.
The fact that the protein-nitrogen-content of the lens does not increase in spite of its steady growth and the proof of proteolytic enzymes in sclerotic and cataractous lenses justify the assumption that a breakdown of protein may occur in the central areas of the lens. The residual molecules which originate in this way should explain the “pathological formation of albuminoid” as well as the shift in the amino acid values.
