Summary
85Sr uptake and ash-weights of the fracture callus of the tibia and of a heterotopic tibia graft in the same animal were measured in mice and followed up to 30 days after grafting and fracture. Growth and uptake of 85Sr was higher in the callus compared to the graft and reached a maximum at about 17 days post fracture. After this period 85Sr uptake of the callus decreased rapidly, whereas in the graft an initial increase of 85Sr uptake was followed by a slow decline after 17 days. Ash-weights of the grafts increased slightly up to day 11 und remained constant thereafter. A low 85Sr uptake of the uninjured tibia between days 2 and 4 post fracture compared to the data of the following period was assumed to be due to a transitory enhanced excretion of 85Sr along with Ca during this early period. Between 9 and 30 days the 85Sr uptake of the uninjured tibia remained unchanged. An influence of fracture repair or graft healing on the mineral uptake of the whole skeleton therefore cannot be confirmed by our data.
Zusammenfassung
Im Tierversuch wurden die quantitativen Veränderungen des Knochenminerals während der Heilung einer Knochenfraktur und einer syngenen, heterotopen Knochentransplantation unter Verwendung von 85Sr vergleichend untersucht.
Der Callus wuchs relativ schnell und nahm im Anfangsstadium bis zum Erreichen seiner größten Ausdehnung relativ viel 85Sr auf. Die höchste Konzentration von 85Sr wurde früher erreicht als das höchste Gewicht bzw. die größte Ausdehnung. Nach Erreichen des höchsten Callusgewichtes wurde zunehmend weniger Mineral angelagert, während gleichzeitig die Resorption zunahm. Infolgedessen wurde der Callus in dieser Phase des „remodelling“ wieder kleiner. Eine Phase ausgeglichener Bilanz, während welcher der Aufbau und die Resorption des Knochenminerals gleich sind, wurde innerhalb des Versuchszeitraumes (30 Tage) nicht erreicht.
Das Gewicht des Transplantats nahm nach einer gewissen Latenzzeit, die zum Anschluß an die Gefäßversorgung des Empfängers nötig ist, nur wenig zu. Während dieser Zeit stieg die Aufnahme von 85Sr deutlich, wenn auch weniger stark als beim Callus an. Auch hier nahm die 85Sr-Ablagerung nach Erreichen eines Maximums wieder ab. Im Gegensatz zum Callus blieb aber von diesem Zeitpunkt an das Gewicht des Transplantats erhalten, d. h., die Resorptionsvorgänge blieben gleich groß wie die Aufbauvorgänge.
Der Heilungsverlauf beim Frakturcallus und beim Transplantat sind gekennzeichnet durch einen deutlichen Anstieg der 85Sr-Aufnahmekapazität bis zu einem Maximum und einer anschließenden Normalisierung und stehen dadurch im Gegensatz zu dem von uns früher beschriebenen gleichförmigen Verlauf der 85Sr-Aufnahme beim allogenen „nicht heilenden“ Transplantat der Maus. Im restlichen Skelet konnte nach Fraktursetzung und Transplantation keine Änderung der 85Sr-Aufnahme festgestellt werden.
