Summary

The tissue changes in osteogenesis imperfecta apparently lie in the collagen and in the glycosaminoglycans. The collagen shows structural development disturbance and incomplete calcification. The tissue glycosaminoglycans are increased. The basis probably lies in a disturbed ATP metabolism (Solomons and Millar, 1973). An increased pyrophosphate concentration explains the decreased calcification and the structural changes. The increased osteolysis could be explained by increased quantities of cAMP.

Zusammenfassung

Die geweblichen Veränderungen liegen bei der Osteogenesis imperfecta offenbar ebenso im Kollagen wie bei den Glykosaminglykanen. Das Kollagen zeigt Reifungsstörungen und eine mangelhafte Verkalkungstendenz. Die Glykosaminglykane des Gewebes sind offenbar quantitativ allgemein vermehrt. Zugrunde liegt wahrscheinlich ein gestörter Stoffwechsel mit einem vermehrten Abbau von ATP, das entweder zu dem energetisch nicht mehr nutzbaren Hypoxanthin degradiert wird oder zu Pyrophosphat und cAMP. Eine erhöhte Pyrophosphatkonzentration erklärt die verminderte Verkalkungstendenz des Kollagens und auch dessen Strukturveränderungen. Die verstärkte Knochenabbaurate könnte auf den vermehrten Anfall von cAMP zurückgeführt werden.
