Summary
The antianginal drug amiodarone (an amphiphilic cationic compound) causes a keratopathy in humans. In the present investigation the cytologic effects of amiodarone on ocular tissues of rats were studied. Ultrastructural alterations, which are typical of human keratopathy and characteristic of drug-induced lipidosis, could be experimentally reproduced in rats by local application of amiodarone. Repeated oral administration of high drug doses induced lipidosis-like alterations in many ocular cell types, particularly in retinal pigment epithelium. It is concluded that amiodarone has the potency to induce generalized lipidosis in rats, as do several other previously investigated amphiphilic cationic drugs. It is tentatively suggested that amiodarone-induced corneal alterations in humans might equally be interpreted as part of a generalized lipidosis.
Zusammenfassung
Das bei Angina pectoris eingesetzte Medikament Amiodaron (Cordaron®) hat beim Menschen verschiedentlich Fälle von Hornhauttrübungen verursacht. In der vorliegenden Arbeit wurde die Wirkung von Amiodaron, das nach seinen physikochemischen Eigenschaften als amphiphile, kationische Verbindung anzusehen ist, auf die Ultrastruktur der Augen von Albinoratten untersucht. Lokale Applikation des Medikaments rief an der Hornhaut der Ratte ultrastrukturelle Veränderungen hervor, wie sie für die Amiodaron-Keratopathie des Menschen und zugleich für die pharmakoninduzierte Lipidose typisch sind. Wiederholte orale Gaben von hohen Dosen des Medikaments verursachten lipidosis-artige Veränderungen an vielen anderen Zellarten des Auges, insbesondere am Pigmentepithel der Retina. Wir schließen aus den vorliegenden Befunden, daß Amiodaron bei der Ratte eine generalisierte Lipidose zu induzieren vermag, so wie es für eine ganze Reihe anderer amphiphiler, kationischer Verbindungen schon früher nachgewiesen worden ist. Zugleich möchten wir den Verdacht äußern, daß die Amiodaron-Keratopathie des Menschen ebenfalls einen Teilaspekt einer pharmakoninduzierten, möglicherweise generalisierten Lipidose darstellen könnte.
