Summary
Yeast cells pre-incubated under anaerobic conditions show an NADH overcompensation during the transition from dormancy to metabolic activity. This is in contrast to the behavior of yeast cells pre-incubated under aerobic conditions. This was demonstrated by Holzer (1960) who found a difference in the rates of NADH-producing and NADH-consuming reactions. Further studies showed that this NADH increase is effectively measurable only in aerobic cultures, while in anaerobic cultures strong interfering reactions occur. It could be shown that the activity measured in anaerobic yeast arose not from oxydation of GAP but from oxydation of endogenous alcohols. The activity is directly dependent upon the presence of alcohol and is not influenced by azide, cyanide or monoiodoacetic acid.
Zusammenfassung
Beim Übergang vom Ruhezustand in den der Stoffwechselaktivität zeigt eine anaerob vorkultivierte Hefe im Gegensatz zu einer aeroben eine NADH-Überkompensation. Diese wurde von Holzer (1960) mit der zeitlichen Differenz zwischen einer NADH-produzierenden und einer NADH-verbrauchenden Reaktion gedeutet. Eingehende Untersuchungen zeigten, daß diese NADH-Zunahme nur bei der aeroben Kultur effektiv meßbar ist, während bei der anaeroben Hefe eine starke Überlagerung zusätzlich auftritt. Es konnte gezeigt werden, daß dieser peak der anaeroben Hefe nicht aus der GAP-Dehydrierung, sondern aus der Dehydrierung endogenen Alkohols stammt. Der peak ist direkt vom Alkohol-Spiegel abhängig und nicht durch Azid, Cyanid oder Monojodessigsäure beeinflußbar.
