Summary
The degree of metachromasia in the normal cornea of humans and pigs was investigated in unfixed cryostat sections with a series of toluidine blue preparations of pH 3.4–4.6. Additional sections stained with Alcian Blue and colloidal iron were used for comparison. The boundary region in both cases is about pH 4.0, an intensity gradient decreasing from outside inwards being seen in the human cornea, but in the opposite direction in pig cornea. Homogeneous staining appears at pH 4.6. While staining with Alcian Blue compares well with complete metachromasia, colloidal iron behaves irregularly and should only be used in conjunction with other staining reactions.
Zusammenfassung
Die Stärke der Metachromotropie der normalen menschlichen Hornhaut und der Schweinecornea wurde an unfixierten Kryostatschnitten in einer Toluidinblau-pH-Reihe von 3,4–4,6 untersucht. Zusätzliche Färbungen mit Alcianblau und kolloidalem Eisen wurden zum Vergleich herangezogen. Der pH-Grenzbereich liegt in beiden Fällen bei pH 4,0. Bei der menschlichen Cornea zeigt sich ein von außen nach innen abnehmend gerichteter Intensitätsgradient, bei der Schweinecornea verläuft er in umgekehrter Richtung. Eine homogene Färbung tritt bei pH 4,6 auf. Während die Alcianblau-Färbung gut mit der kompletten Metachromasie vergleichbar ist, zeigt die kolloidale Eisenfärbung ein inkonstantes Verhalten; sie sollte nur in Verbindung mit anderen Farbreaktionen angewandt werden.
