Summary
The influence of oxyphenbutazone on the carbohydrate metabolism of calf and bovine lenses was studied in vitro. The activities of 9 enzymes as well as the concentrations of 7 substrates were determined. The content of ATP and ADP was decreased and that of AMP increased after a 24 h culture in a medium containing 10−2 m oxyphenbutazone. Content of glucose, glucose-6-phosphate and fructose was decreased. On the other side, oxyphenbutazone caused an increase of fructose-1,6-diphosphate. The significance of the results obtained could be proved.
The activities of glucose-6-phosphate dehydrogenase and pyruvate kinase were decreased by the substance. Addition of 10−4 m oxyphenbutazone to the medium showed no important effect; in calf lenses only a small but significant increase in ATP content was found. A possible mechanism of the effect on the energy balance is discussed.
Zusammenfassung
An Kälber- und Rinderlinsen wurde der Einfluß von Oxyphenbutazon auf den Kohlenhydratstoffwechsel in vitro untersucht. 9 Enzymaktivitäten und 7 Substratkonzentrationen wurden bestimmt. Nach 24stündiger Kultivierung mit 10−2 m Zusatz von Oxyphenbutazon zur Nährlösung verringerten sich mehrere Substratkonzentrationen signifikant. Dies betraf vor allem die energiereichen Adenosinphosphate, ferner den Gehalt an Glucose, Fructose und Glucose-6-phosphat. Die Glucose-6-phosphatdehydrogenase und die Pyruvatkinase zeigten erniedrigte Aktivitäten. Ein Zusatz von 10−4 m Oxyphenbutazon zur Nährlösung blieb ohne erkennbare Wirkung, lediglich bei Kälberlinsen fand sich eine geringe, jedoch signifikante Zunahme im Gehalt an Adenosintriphosphat.
