Summary
Single s.c. applications of N-nitrosobis(2-hydroxypropyl)amine (BHP) caused carcinogenic effects in adult Syrian golden hamsters at doses as low as 1/40 of the LD50. The tumor spectrum decreased at lower doses, which affected the pancreas more strongly than the liver, whereas higher doses exhibited a more ponounced hepatocarcinogenicity and also induced tumors in the respiratory tract, gallbladder, kidney, and vagina. These results demonstrated a potent pancreatic carcinogenic effect of BHP at the lowest dose level. Hence, the presence of BHP in the environment, even in small quantities, should be regarded as a potential hazard to human health.
Zusammenfassung
Einmalige s.c. Applikation von N-Nitrosobis(2-hydroxypropyl)amin (BHP) zeigte in Dosen bis zu 1/40 der LD50 eine karzinogene Wirkung bei erwachsenen Syrischen Hamstern. Pankreas und Leber waren die Hauptorgane für Tumorinduktion. Das Spektrum des Tumors verkleinerte sich in Abhängigkeit von der BHP-Konzentration und war minimal in der kleinsten Dosis, bei der das Pankreas am stärksten betroffen war. Dagegen waren hohe Dosen mehr leberspezifisch und induzierten Tumoren auch in dem Respirationstrakt, in der Gallenblase, der Niere und der Vagina. Wegen der karzinogenen Wirkung der kleinsten Dosis von BHP insbesondere auf das Pankreas, muß das Vorkommen dieses Karzinogens, sogar in kleinsten Mengen, in der Umwelt als gesundheitsschädlich für den Menschen angesehen werden.
