Nachdem der einwandfreie Nachweis der Heterokaryose bei Fusarium oxysporum bisher nicht gelungen ist, waren Untersuchungen notwendig, ob die Voraussetzungen für die Bildung von Heterokaryonten, ihre experimentelle Synthese und Analyse erfüllt sind.

In einer vorangegangenen Arbeit wurde über die Kernverhältnisse im vegetativen Mycel von Fusarium oxysporum berichtet; bei den vorliegenden Untersuchungen wurden Beobachtungen über die Entwicklung von Anastomosen, ihre Formen und deren relative Häufigkeit, ihr Auftreten unter bestimmten Umweltbedingungen und in verschiedenen Entwicklungsstadien des Mycels durchgeführt.

In mehreren Versuchsreihen mit fünf verschiedenen Erregerherkünften wurde festgestellt, daß die auf künstlichen Nährböden erwachsenen Conidien während oder kurz nach der Keimung keine Neigung zur Bildung von Anastomosen besitzen. Conidien von Fusarium oxysporum f. callistephi aus Sporodochien an erkrankten Pflanzen bildeten nur in nährstofffreiem Keimmedium zwischen den Sporenzellen oder deren Keimschläuchen Anastomosen.

Hyphenverbindungen verschiedener Art traten zu über 95% unter Beteiligung älteren Mycels auf. Meist wurden kurze, einzellige Anastomosen an eng nebeneinanderliegenden Hyphen gebildet, wobei die Zellverbindungen nach beiden Seiten offenbleiben.

Mikro- und Makroconidien von Fusarium oxysporum sind homokaryotisch auch nach Bildung an vielkernigem Mycel. Die Conidienträger können mit der Basiszelle in offener Verbindung bleiben und unterliegen in Koordination mit der Conidienbildung fortgesetzter Einwanderung von Kernen.

Die Untersuchungen zeigen, daß die Voraussetzungen für Heterokaryosebildung bei Fusarium oxysporum in künstlicher Kultur nicht in idealer Weise erfüllt sind. Obgleich Heterokaryose möglich ist, dürfte sie durch die Veränderlichkeit der Kernverhältnisse unstabil bleiben. Die geringe Paarungsneigung von Conidien bzw. von jungem Mycel auf nährstoffhaltigen Substraten, der offenbar geringe Kernaustausch auch im älteren Mycel erschweren den Nachweis der Heterokaryose durch Hyphenspitzenisolierungen, so daß die erfolglosen Bemühungen verschiedener Autoren dadurch eine Erklärung finden.

As no definite proof of heterokaryosis of Fusarium oxysporum has yet been found, investigations were necessary to see whether the conditions for the development of heterokaryons, their synthesis and analysis are in fact fulfilled.

In a previous publication a report was given on the relation of nuclei in the vegetative mycelium of Fusarium oxysporum; in the present investigations observations were carried out on the development of anastomoses, their forms, on their relative occurrence, and on their appearance under definite environmental conditions as well as in different stages of mycelium development.

In several series of tests with five different strains it was noticed that conidia, which grew on artifical nutrient media, possessed no tendency to form anastomoses during or shortly after germination. Conidia of Fusarium oxysporum f. callistephi from sporodochia of diseased plants, formed anastomoses between themselves or their germ bubes only if grown on a nutrient free germination medium.

Over 95% of hyphal fusions of different kinds occurred in older mycelia. Most commonly, short, single-celled anastomoses were formed between hyphae that were lying close together, in which case the cell connections on both sides remained open.

Micro- and macroconidia of Fusarium oxysporum are homocaryotic even if formed on a multinucleate mycelium. The conidiophores can persist in an open connection with the basal cell and remain in coordination with the continual immigration of nuclei into the developing conidia.

The investigations show that the conditions for heterokaryosis of Fusarium oxysporum are not ideal in an artificial nutrient-medium culture. Though heterokaryosis is possible, it could remain unstable due to changes in the number of nuclei. As the numbers of anastomoses between conidia and young mycelia on an artificial nutrient-medium are low, and, also, as apparently slight nuclear exchange between older hyphae complicate the proof of heterocaryosis through the isolation of hyphal tips, the unsuccessful efforts of different authors can be explained.
