The influence of research-material on the ATP-level of Rhodospirillum rubrum in anaerobic dark culture

Summary

When dark cultures of Rhodospirillum rubrum were shifted from aerobic to anaerobic cultural conditions, the ATP-level (within the cells) changed dependent on the growth conditions:

In presence of substrate and (NH4)2SO4 the ATP-concentration went through a pronounced minimum and maximum before the new plateau was reached. In absence of substrate no such oscillation or at best a slight irregularity could be observed. The new ATP-level was maintained or at least its decline slowed down by means of reserve-material (synthesized during aerobic cultivation).

Exogenous substrate did not enlarge the ATP-pool under anaerobic conditions in the dark, rather it may enhance its decline. The ATP-pool within the cells was increased only with fructose as substrate and only after a cultivation of 2 days.

After the partial pressure of oxygen had fallen to zero, the ATP-level as a rule reached the minimum in less than 3 min and arrived at its new level within 5 to 60 min.

Zusammenfassung

Beim Übergang von aerober zu anaerober Dunkelkultur wurde der ATP-Spiegel in den Zellen von R. rubrum in Abhängigkeit von den Sachstumsbedingungen neu einreguliert:

In Kulturen mit Substrat und (NH4)2SO4 erhielten wir ein ausgeprägtes Minimum und Maximum bevor sich ein neuer ATP-Spiegel einstellte; in Kulturen ohne Substrat traten keine oder höchstens geringe Schwankungen auf.

Durch Speicherstoffe wurde der neu eingestellte ATP-Spiegel aufrecht gehalten oder der Abfall zumindest verlangsamt.

Exogenes Substrat erhöhte den ATP-Pool anaerob im Dunkeln nicht, es konnte sogar den Abfall beschleunigen. Nur mit Fructose als Substrat wurde, aber erst nach 2 tägiger Bebrütung anaerob im Dunkeln, der ATP-Pool in den Zellen erhöht.

Nach Abfall des Sauerstoffpartialdrucks auf Null erreichte der ATP-Spiegel in der Regel das Minimum in weniger als 3 min und hatte sich in 5–60 min auf einen neuen Wert eingestellt.
