Summary
After injection of 3H-thymidine autoradiographic studies were carried out to determine the uptake, localization, time course, and degree of labeling in the germinal centers of Peyer's patches and mesenteric lymph nodes of adult mice. In some of these experiments a prednisolone-derivative “Solu-Decortin-H”® (Merck) was administered. Besides intact cells nuclear fragments, known as “tingible bodies”, were also taken into account. The ratio of tingible bodies to intact cells remained approximately constant (20∶100). Ten per cent of the labeled cells are destroyed within 24 hours and appear as labeled tingible bodies. The rate of cellular breakdown within the germinal center is not higher than in other lymphatic organs. Therefore, destruction of the young cells with high mitotic rate within the center (“germinoblasts”) cannot be their predominant fate. The conclusion reached from the experiments carried out is that the germinal centers are sites of production of mobile cells that readily leave their site of formation but do not enter the peripheral part of the secundary nodule.
Zusammenfassung
In Sekundärfollikeln von Peyerschen Plaques und darmnahen Mesenteriallymphknoten erwachsener Mäuse wurden nach Gaben von 3H-Thymidin seine Aufnahme und Lokalisation sowie zeitlicher Verlauf und Grad der Zellmarkierung autoradiographisch untersucht. Neben den intakten Zellen wurden die tingiblen Körperchen nach Anzahl und Beladung erfaßt. Ein Teil der Tiere erhielt zusätzlich ein Prednisolon-Präparat (Solu-Decortin-H ® Merck) injiziert. Das Zahlenverhältnis der tingiblen Körperchen zu den intakten Zellen ist nahezu konstant 20∶100. Von den radioaktiv markierten Zellen werden 10% innerhalb 24 Std in Kerntrümmer verwandelt. Die Absterberate der Zellen im Zentrum von Sekundärfollikeln ist nicht größer als in anderen lymphatischen Organen. Das normale Schicksal der Germinoblasten kann nicht im Zelluntergang am Ort gesehen werden. Die Follikelzentren sind Bildungsorte mobiler Zellen, die rasch ausgeschleust werden. Eine Überwanderung in den Lymphocytenwall findet nicht statt.
