Growth and fermentation of a new isolate of Clostridium aceticum wieringa

Summary

  1. 1.

    A mesophilic Clostridium was isolated by enrichment culture containing pond-mud, mineral salts, and vitamins under an atmosphere of 75% H2 and 25% CO2 at 28°C. This Clostridium synthesized appreciable quantities of acetic acid from molecular hydrogen and carbon dioxide.

  2. 2.

    Acetate and formate were produced by the fermentation of fructose, glutamate, lactate and pyruvate. In addition to one of these organic substrates, vitamins, amino acids and some trace-elements were required for growth. This Clostridium is strictly anaerobic; however, in the presence of 4 μg cobalt/ml it even grows without anaerobic precautions. The temperature optimum lies between 37 and 45° C.

    This new strain of Clostridium aceticum is differentiated from that isolated by Wieringa (1936) by a more restricted substrate spectrum and by the formation of formic acid in addition to acetic acid.

    Aus dem Schlamm eines Teiches wurde in einer vitaminhaltigen. Mineralsalzlösung unter einer Atmosphäre von 75% H2 und 25% CO2 bei 28°C ein mesophiles Clostridium angereichert und isoliert, das aus Kohlendioxyd und molekularem Wasserstoff große Mengen Essigsäure bildet.

    Das Clostridium vergärt Fructose, Glutamat, Lactat oder Pyruvat zu Acetat und Format. Es benötigt zum Wachstum eines dieser Substrate und zusätzlich Aminosäuren, Vitamine und Spurenelemente. Es ist streng anaerob, wächst jedoch in Gegenwart von 4 μg Kobalt/ml auch ohne Anaerobenverschluß. Das Temperaturoptimum liegt zwischen 37 und 45° C.

    Der neuisolierte Stamm unterscheidet sich von dem von Wieringa (1936) isolierten Clostridium aceticum durch den engeren Substratbereich und die Bildung von Ameisensäure neben Essigsäure.
