Während intracellulärer Ableitung aus Nervenzellen der motorischen Rinde der Katze wurde die Wirkung frequenter elektrischer Reizungen der Formatio reticularis mesencephali untersucht.

Das Membranpotential vergröβert sich während der Reticularisreizung nach etwa 200 msec um 2–8 mV in Richtung der Hyperpolarisation.

Abhängig von der Höhe des Membranpotentials vor der Reizung kann es zu verminderter Entladungsfrequenz kommen oder zu kompletter Hemmung der Spontanaktivität (20% der untersuchten Zellen).

Lediglich 12,5% der beobachteten Nervenzellen zeigten eine Zunahme der Entladungsfrequenz oder eine Latenzverkürzung bei Zellantworten nach Reizung spezifischer Thalamuskerne.

Bei gleichzeitiger Reizung spezifischer oder unspezifischer Thalamuskerne während des arousal-Reizes wird die Zellantwort auf die letztere häufiger unterdrückt als die auf Reizung des spezifischen Thalamuskernes (V.o.p.).

Bei Primärantworten auf frequente Reizung des spezifischen Thalamuskernes verlängert sich die Latenz der Zellentladung durch die begleitende Polarisationszunahme durch Reticularisreizung um 2–4 msec, sofern der Thalamusreiz überhaupt noch beantwortet wird.

Während bei einer 300/sec-Reizung der Formatio reticularis keine postsynaptischen Potentiale (PSP) zu erkennen sind, erscheinen bei 100/sec-Reizen PSP, die excitatorischen postsynaptischen Potentialen (EPSP) entsprechen. Typische isolierte inhibitorische postsynaptische Potentiale (IPSP), die der Reizfrequenz folgten, wurden nicht beobachtet.

Werden während der Reticularisreizung EPSP durch gleichzeitige niederfrequente Thalamusreizung ausgelöst, so vermindert sich deren Amplitude — verglichen mit solchen vor der Reticularisreizung — bei annähernd gleicher Polarisation zunächst nicht, bis erst bei höherer Membranpolarisation kleinere EPSP auftreten.

Im Gegensatz zu den Effekten nach Reticularisreizung kommt es nach frequenter Reizung unspezifischer Thalamuskerne eher zu einer Depolarisation der Membran und die Latenz der Reizantworten nach Reizung spezifischer Thalamuskerne verkürzt sich.

Zeitliche Übereinstimmungen zwischen Membranpotentialveränderungen der Neurone und beschriebenen Gleichspannungsänderungen der Rindenoberfläche werden erwähnt: Während des Beginns einer Reticularisreizung erscheint in Gleichspannungsableitungen eine sogenannte „positive Nase“, während der keine oder leicht depolarisierende Membranpotentialveränderungen zu beobachten waren. Nach 300 msec kommt es zu einer Verschiebung des Gleichspannungspotentials in negative Richtung, die während der Reizung anhält und sie überdauert. Dem entspricht zeitlich in etwa der Beginn der Polarisationszunahme der Zellen.

Mögliche synaptische und humorale Mechanismen der Befunde werden diskutiert, besonders eine Depolarisation apikaler Dendriten und präsynaptischer Fasern mit konkurrierender Blockade für nachfolgende Erregungen, Summation der Repolarisationsphasen axo-dentritischer EPSP bei kurzen Reizintervallen, eine Unterdrückung der Spontanrhythmen und der damit einhergehenden synchronen Erregbarkeitsschwankungen oder eine unterschwellige antidrome Erregung.
