Computer controlled large scale production of α-interferon by E. coli

Abstract

Recombinant E. coli expressing alpha-interferon is used for production in a high cell density fed-batch process on 1000 l scale. Computer recorded online fermentation parameters include dissolved oxygen, offgas analysis, glucose feed rate, pH, temperature, stirrer speed, pressure, air and oxygen feed rates, volume and turbidity. Setpoints of temperature, stirrer speed, aeration rates and glucose feed rates are computer controlled. The glucose consumption rate is calculated indirectly from the offgas analysis. The online turbidity signal is used to trigger a temperature ramp. The dissolved oxygen is controlled between allowed limits by adjusting stirrer speed, aeration and pure oxygen feed rates and temperature. The hardware, sensor and control schemes are described and explained.
