Summary
The aim of this study was to investigate the morphologic changes of the submandibular gland during a normal circadian secretory cycle. Untreated male Wistar rats, weighing 250 ± 20 g were sacrificed at 10, 16, 20, 24 and 4 hrs. At each time point four animals were used. Pieces of the submandibular gland were fixed in glutaraldehyde and osmium and processed for electron microscopy. 1 μ thick epon sections were stained with Toluidine Blue. Randomly chosen areas of glandular parenchyma were photographed and morphometrically evaluated using a square lattice. Test points lying on the following acinar structures were counted: Nuclei; secretory parts (i.e. cytoplasmic areas occupied mainly by secretory granules); ergastoplasma parts (i.e. cytoplasmic areas occupied mainly by rough endoplasmic reticulum and therefore heavily stained with Toluidine Blue); lumina (Fig.2).
The volume density of the secretory part of the acinar cells is maximal at 20 hrs. at the onset of food intake, and minimal at 4 hrs., at the end of the eating period. The volume density of the ergastoplasma part is maximal at 4 hrs. and minimal at 20 hrs. The total volume of the acinus however is maximal at 24 hrs. and smallest at 10 hrs., suggesting a predominating increase of ergastoplasma and newly formed secretory granules in the first hours of the eating period (Fig. 11 and 12).
As seen with both, the light and electron microscope, at 20 hrs. the acinar cells are filled up with secretory granules, which often combine to form larger secretory vacuoles. The rough endoplasmic reticulum is scattered throughout the cell between the secretory granules, with only a small part forming compact areas at the cell base. The nuclei appear dark and condensed, being often squeezed and deformed by secretory granules and vacuoles. The Golgi areas are small (Fig.3 and 9).
At 4 hrs. the secretory granules are reduced in number and are mainly located at the apical part of the acinar cell, whereas the main part of the cytoplasma is occupied by compact rough endoplasmic reticulum. The nuclei are enlarged, round and show distinct nucleoli. The Golgi areas are large; the acinar lamina and intercellular spaces are widened and show microvilli and many infoldings of the plasma membrane (Fig. 5 and 10).
Therefore, the submandibular gland of normally fed untreated rats shows a timestructure, i. e. a day-time dependant morphology, caused by rhythmic changes in production, storage and secretion of secretory material.
Zusammenfassung
Ziel dieser Untersuchung war es, die morphologischen Veränderungen der Glandula submandibularis wahrend eines normalen Sekretionscyclus, also unter physiologischen Bedingungen zu studieren. Zu diesem Zwecke wurden jeweils vier unbehandelte, männliche Wistar-Ratten mit einem Gewicht von 250 ± 20 g während einer normalen Tag-Nacht-Periode zu folgenden Zeitpunkten getötet: 10 Uhr, 16 Uhr, 20 Uhr, 24 Uhr and 4 Uhr. Mehrere Stücke von der Glandula submandibularis wurden sofort entnommen, in Glutaraldehyd und Osmium fixiert und in Epon eingebettet. Nach Färbung von 1 μ dicken Semidünnschnitten mit Toluidinblau wurden zufällig ausgewählte Bezirke des Drüsenparenchyms photographiert und mittels eines Testpunktgitters morphometrisch ausgewertet. Dabei wurden Testpunkte auf folgenden Strukturen eines Acinus registriert: Kerne; Sekretanteil (= Cytoplasmabereich, der hauptsäehlich von Sekretgranula eingenommen wird); Ergastoplasmaanteil (= Cytoplasmabereich, der hauptsächlich von rauhem, endoplasmatischen Reticulum eingenommen wird und sich mit Toluidinblau homogen anfärbt); Lumen (Abb. 2).
Die Volumsdichte des Sekretanteiles der Acini ist am größten um 20 Uhr, d. h., zu Beginn der Nahrungsaufnahme, und am kleinsten um 4 Uhr, das ist am Ende der Freßperiode. Die Volumsdichte des Ergastoplasmaanteiles der Acini hat hingegen ihr Maximum um 4 Uhr und ihr Minimum um 20 Uhr. Das Acinusvolumen ist am kleinsten um 10 Uhr und am größten um 24 Uhr (Abb. 11 and 12).
Wie licht- bzw. elektronenmikroskopische Untersuchungen zeigen, sind die Acinuszellen um 20 Uhr dicht angefüllt mit Sekretgranula, die öfter zu größeren Sekretvacuolen konfluieren. Ein großer Ted des rauhen endoplasmatischen Reticulums liegt verstreut innerhalb der Acinuszellen zwischen den Sekretgranula, während die kompakten Membranpakete auf schmale Zonen an der Zellbasis beschränkt sind. Die Zellkerne erscheinen dunkel und kondensiert, und sind oft durch Sekretgranula und Sekretvacuolen eingedellt. Die Golgi-Komplexe sind klein und unauffällig (Abb. 3 and 9).
Um 4 Uhr sind die Sekretgranula auf den apikalen TO der Acinuszelle beschränkt, während der größte Teil des Cytoplasma von kompaktem, rauhen, endoplasmatischen Reticulum eingenommen wird. Die Zellkerne sind groß, rund und zeigen deutliche Nucleolen. Die ausgedehnten Golgi-Komplexe treten deutlich hervor. Die Acinuslumina und die Intercellularräume sind erweitert, die Cytoplasmamembranen zeigen Mikrovilli und zahlreiche Einfaltungen (Abb.5 and 10).
Die Glandula submandibularis von normal gefütterten, unbehandelten Ratten weist somit eine ausgeprägte Zeitstruktur auf: Der jeweiligen Tageszeit entspricht ein charakteristisches morphologisches Erscheinungsbild, welches die rhythmisch wechselnde Tätigkeit der Drüsenzelle, nämlich Produktion, Speicherung und Abgabe von Sekret, widerspiegelt.
