Zusammenfassung
Das venöse Hirnblut von Kaninchen wurde während der elektrischen Reizung des aktivierenden Retikularsystems dialysiert. Das Dialysat wurde an einem freibeweglichen Receptortier auf seine Wirkung geprüft. Das Verhalten wurde gefilmt und mit Hilfe eines Kinesimeters analysiert. Die gleichzeitig registrierte elektrische Hirnaktivität wurde mit einem automatischen Frequenzanalyser ausgewertet. In einer Reihe von Kontrollversuchen wurde Dialysat von Spendertieren, bei welchem nur eine Scheinreizung des Retikularsystems durchgeführt worden war, normalen Receptoren injiziert.
Die Receptoren, welche 20 ml Dialysat von einem stimulierten, wachen Spender erhielten, wiesen schon in den ersten 5 min nach Injektion ein aktiviertes Spontanverhalten auf, meistens mit vielfältigen, rhino-diencephalen Symptomen. Parallel dazu zeigte das EEG ein typisches Wachbild mit Abnahme der langsamen delta-Rhythmen, Desynchronisierung der Aktivitäten im Cortex und reichlichen theta-Aktivitäten im Hippocampus.
Kontrollreceptoren, welche Dialysat von nicht-stimulierten, jedoch infolge der Dialyse leicht aktivierten Spendern erhalten hatten, wiesen ebenfalls eine leichte Aktivierung des Verhaltens und der elektrischen Hirnaktivität auf, aber meistens ohne ausgeprägte rhino-diencephale Symptome.
Diese Befunde lassen vermuten, daß die Aktivierung des Receptors, vor allem seine rhino-diencephale Symptomatik, nach Injektion von Dialysat eines stimulierten Spenders auf der Wirkung spezieller, aus dem Blut extrahierter „Faktoren“ beruht.
Summary
Dialysis of cerebral venous blood was performed during repetitive stimulation of the reticular activating system of the rabbit. The dialysate obtained in this way from an awakened donor was injected intravenously into a normal animal (recipient). Alterations in behaviour and EEG were recorded and analysed (film, kinesimetry, automatic frequency analysis of the EEG). In control experiments dialysate was injected into normal recipients from animals in which sham-stimulation of the activating reticular system had been performed.
Within 5 min after injection of 20 ml of dialysate from an alert donor, recipient animals revealed activated behaviour with various rhino-diencephalic symptoms (explorative behaviour, sniffing, lapping, grooming). Concurrently, a typical arousal pattern appeared in the EEG, with desynchronised activities in the cortex and abundant theta rhythms in the hippocampus. Control animals which received dialysate from sham-stimulated donors also showed activation of behaviour and electrical brain activity, but less pronounced and mostly without rhinencephalic symptoms.
These results suggest that activation of rhino-diencephalic type observed in the recipient after injection of dialysate from a stimulated donor is due to some factors released in the donor's blood and extracted by dialysis.
