Summary
The host-parasite relationship in Fasciola hepatica infection of albino rats were studied in male SPF Wistar-rats. After infection with 35 metacercariae some serum- and blood parameters were examined for 10 weeks especially in correlation to the liver migration and fertility of the parasites.
28 days after infection the first few flukes could be already isolated from the main bile duct. Between the 32nd and 36th day p.i. nearly 60% of the flukes had entered the biliary system. In five rats only one immature fluke was found in the liver parenchyma 52 days p.i. The prepatent period was at least 40 days.
In the acute phase of infection there were characteristic increases of GLDH, SDH, GPT, GOT, leucocytes and eosinophilic leucocytes. The maximum levels of these parameters were directly correllated with the main invasion of the flukes into the biliary system. The activity of GLDH especially was most marked elevated. With the patency of the infection all values except of Hb and LAP showed an obvious tendency to normalisation. The alterations in LAP, ChE and bilirubin were only slight without showing a continuous development.
10 weeks after infection only low significant differences in comparison to controls could be demonstrated for the average values of SDH, LAP and leucocytes whereas the Hb-values and eosinophilic leucocytes remained highly significant different.
Zusammenfassung
In Untersuchungen über das Wirt-Parasit-Verhältnis bei der Fasciola hepatica-Infektion der Albinoratte wurden männliche SPF Wistar-Ratten mit 35 Metacercarien infiziert. Im Verlauf einer 10wöchigen Beobachtungszeit wurden die Veränderungen einer Anzahl Serum- und Blutparameter untersucht und insbesondere mit der Wanderung und Reifung der Parasiten in Beziehung gebracht.
Bereits 28 Tage p.i. konnten vereinzelte Leberegel aus dem ampullenförmig erweiterten Hauptgallengang isoliert werden. Zwischen dem 32. und 36. Tag p.i. war mit ca. 60% der größte Anteil der Parasiten in die Gallengänge eingewandert. 52 Tage p.i. wurde nur noch ein juveniler Parasit im Leberparenchym von 5 Ratten gefunden. Die Präpatenz betrug mindestens 40 Tage.
Von den ausgewählten Indikatoren zeigten GLDH, SDH, GPT und GOT sowie Leukozyten und eosinophile Granulozyten charakteristische, hoch signifikante Anstiege in der akuten Phase der Parasitose. Die maximalen Erhöhungen der Werte dieser Parameter fielen in den Zeitraum der hauptsächlichen Gallengangsbesiedlung, wobei insbesondere die GLDH mit erheblichen Anstiegen reagierte. Mit Beginn der Patenz konnte bis auf die Hb- und LAP-Werte eine Tendenz zur Normalisierung beobachtet werden. Die Veränderungen von LAP, ChE und Bilirubin waren nur mäßig ausgeprägt und ließen keine kontinuierliche Entwicklung hinsichtlich der statistischen Beurteilung erkennen.
Zum Versuchsende 10 Wochen p.i. waren nur noch bei SDH, LAP und der Gesamtzahl der Leukozyten leicht signifikante, dagegen bei dem Hb-Gehalt und den eosinophilen Granulozyten hoch signifikante Unterschiede festzustellen.
